Mesona chinemis Benth., also known as Xiancao immortal grass, grass jelly, immortal freeze, salary grass, for the Labiatae Xiancao. an annual herb, is an important drug edible plant resources in the East. Relevant literature research shows that, informed research focused on polysaccharides, some of the recent researchs related to flavonoids and phenolic acids. Modern pharmacological studies have shown that Mesona chinemis Benth. has many medicinal value, such as anti-hypoxia, anti-oxidation, clear free radicals, decrease blood sugar, antibacterial, et. Now it is mainly used in the food and the health food field. There are many medicinal material sources without a uniform quality standard. It is inconvenient to the future acquisition, the development and the quality control. The research object of this paper is the aboveground parts of Mesona chinemis Benth..The aim of the research is to study the quality control of Mesona chinemis Benth., providing science basis and foundation for further development and application.This paper mainly research on related literatures, systematic pre-experiment, extraction technology and the determination of total flavonoids, the fingerprint technology of Mesona chinemis Benth. and its quality evaluation.Literature Research:In this paper, germplasm resources and the distribution, biological form and biological characteristics, identification, chemical constituents, pharmacological and toxicological study, application research and research prospects of Mesona chinemis Benth. are summarized, also the research situation of fingerprint of traditional Chinese medicine, including the conception and classification, the fingerprint requirements and application, the application of Chemical pattern recognition, in order to further design and implementation of the experimental scheme provides theory basis.Systematic pre-experiment:The components in Mesona chinemis Benth. contain polysaccharide, flavonoids, phenols and triterpenoid, etc. which based on systematic pre-experiment, chemical identification and the extracts by different polarity solution (petroleum ether, chloroform, ethyl acetate, butanol, methanol, ethanol and water). Besides polysaccharide, flavonoids especially soluble ones are mostly among these components. Combined with the result of research on pharmacology and pharmacodynamics of Mesona chinemis Benth., the research content of the extraction technology in this paper contains mainly aim at flavonoids compounds.Extraction technology and determination of total flavonoids:Single factor exploration determined single factor level, the five factors are as follows:concentration of ethanol, extraction temperature, extraction time, amount of solvent, extraction times. Select factor and level for orthogonal design L9 (3.4), study the three factors:extraction time, amount of solvent and extraction times, and how they effect the extraction efficiency. Finally determine the optimum extraction factor of Mesona chinemis Benth.:ten times 50% ethanol, extraction temperature is 100℃, extract two times, each 45 min. In order to regulate the quality of Mesona chinemis Benth., the leaching rate of total flavonoids should not be less than 4.0%.The fingerprint technology:This method mainly study on the preparation of test solution, chromatography conditions selection and methodology investigation. Chromatography conditions, including mobile phase gradient, .detection wavelength, detection time, different types of HPLC columns, different types of acid and their various concentration, flow rate and column temperature,.were investigated. The HPLC fingerprint of Mesona chinemis Benth. was established, preliminarily 10 characteristic peaks were selected and the similarity among 34 batches of Mesona chinemis Benth. from different aera have been determined. In addition, the results of cluster analysis showed that samples from different areas can be divided into three categories.Quality evaluation:The the content of total flavonoids and the fingerprint was used as reference to evaluate the quality of Mesona chinemis Benth. in different collecting time and processing methods, to provide scientific basis for the construction of GAP base. As a result,.suitable harvest time is the afternoon of September and October in summer, from 12:00 to 17:00, the best time is about 16:00.. Primary processing methods need to be further validation of the pharmacological efficacy.
