Objective:The study using the technology of RT-PCR compared with the expression level of CLCFl gene of POP and Kidney Yin deficiency group、POP and Kidney Yang deficiency group、POP and other Kidney deficiency group、POP and non-Kidney deficiency group and normal group for the purpose of verifying the relationship between expression of CLCFl gene and postmenopausal osteoporosis with Kidney Yin deficiency. So that the genomic mechanism of postmenopausal osteoporosis with Kidney Yin deficiency be investigated.Methods:Postmenopausal osteoporosis subjects were randomly selected in fuzhou,by questionnaire survey.DXA (Dual energy X-ray bone mineral density testing) and syndrome differentiation of traditional Chinese medicine.They were randomly divided into four groups: POP and Kidney Yin deficiency group (n=32), POP and Kidney Yang deficiency group (n=26),POP and other Kidney deficiency group (n=43),POP and non-Kidney deficiency group (n=30), in addition, the health postmenopausal women as control group were included (n=30). The blood samples were gathered, and then Leukocytes in peripheral blood of each person was insolated by red blood cell lysis buffer.Extract white blood cells total RNA with Trizol reagent, and then test the quality of RNA, do the reverse transcription. Finish each group CLCF1gene and GAPDH gene RT-PCR reactions, rendering the DNA standard curve of serial dilutions, and then get the relative content of the gene, through the GAPDH gene concentration divided by CLCF1gene concentration. Finally, through SPSS statistical method, the expression of each group CLCF1gene were compared with each other.Results:The expression of CLCF1gene concentration after adjusting:POP and Kidney Yin deficiency group:0.0171±0.0109; POP and Kidney Yang deficiency group:0.0867±0.2881; POP and other Kidney deficiency group:0.1776±0.4895; POP and non-Kidney deficiency group:0.0672±0.2539; normal group:0.1642±0.4290。 Compared with POP and Kidney Yang deficiency、POP and other Kidney deficiency、POP and non-Kidney deficiency, the expression level of CLCF1gene of POP and Kidney Yin deficiency levels was not statistically significant (P>0.05), compared with the normal group, the expression level of CLCF1of POP and Kidney Yang deficiency group significantly reduced (P<0.01); Compared with POP and other Kidney deficiency、POP and non-Kidney deficiency、normal group, the expression level of CLCF1gene of POP and Kidney Yang deficiency levels was not statistically significant (P>0.05); Compared with POP and non-Kidney deficiency、normal group, the expression level of CLCF1gene of POP and other Kidney deficiency levels was not statistically significant (P>0.05); Compared with normal group, the expression level of CLCF1gene of POP and non-Kidney deficiency levels was not statistically significant (P>0.05)Conclusions:Compared with each group CLCF1gene expression by RT-PCR technology, which only CLCF1gene expression level of POP with Kidney Yin deficiency group was lowed significantly than the normal group (P<0.01),the remaining groups CLCF1gene expression were not statistically significant (P>0.05). The result was consistent with detection results of gene microarray in the preliminary topics, And it verified some of the reliability of the results of gene chip expression.It’s comfirmed that there was some association between Postmenopausal osteoporosis with Kidney Yin deficiency and the expression level of CLCF1gene. |
