The Clinical Study On TNF-α,TGF-β And ADAM10of Children Primary Nephrotic Syndrome By Treatment Of Tonifying The Kidney And Invigorate The Circulation Of Blood

Objective:by observing the change of TNF-α TGF-β、ADAM10before and after intervening Primary Nephrotic Syndrome kids, in order to expound pathogenesis better of PNS and the treat mechanism of Chinese Traditional Medicine ShenKang ling.Method:60kids with PNS were randomly divided into test group(A,Shenkangling+prednisone)with30cases and control group (B,prednisone+statin lipid-lowering dr-ugs+anticoagulant drug)with30cases. Another to select the six cases of healthy ch-ildren of the medical center as a healthy control group (normal group, Group C).Coll-ect intravenous blood10-15ml from each child in8:00am, before the treatment and2weeks after urinary protein is negative. Test TNF-α、TGF-β、ADAM10by ELISA method, Statistical analysis was performed by SPSS13.0system software.Result:①Prior treatment, the levels of TNF-α、TGF-β、ADAM10of A and B are apparently higher than C (P=0.000).②fter3-month-treatment, the levels of TNF-α、 TGF-β、ADAM10are apparently lower than before (P=0.000),and the levels TNF-α、 ADAM10of A are apparently lower than B (P=0.009).③fter3-month-treatment, th-e levels of plasma fibrinogen, cholesterol, triglyceride, low-density lipoprotein are app-arently lower than before (P=0.000),and the levels of serum albumin, high-density li p-oprotein are apparently higher than before (P=0.000). However, the levels of choles t-erol, triglyceride have no difference with prior treatment (P=0.053, P=0.072). After3-month-treatment, comparing A with B, A is better than B in reducing plasma fibrino-gen, cholesterol, triglyceride, low-density lipoprotein (P=0.000, P=0.000, P=0.000, P=0.001),as well as in improving serum albumin, high-density lipoprotein (P=0.004, P=0.000).④After3-month-treatment, the remission rate of A and B is separately96.7%and90.0%, there is no statistics difference(P=0.351)Conclusion:the levels of TNF-α、TGF-β、 ADAM10are abnormal in PNS serum detec-tion, it just illustrates that TNF-α、TGF-β、 AD AM10take part in pathogenesis in PN S:MCs release cytokines by autocrine and paracrine, and precipitate the proliferation, apoptosis of MCs, lead to glomerulus fibrosis. It can remarkablely reduce the levels of TNF-α、TGF-β、 AD AM10by applying the method of tonifying the kidney and invi-gorating the circulation of blood, reduce fibrinogen and blood fat, improve serum alb- umin, in order to adapt the inter environment of high fat and hypercoagul ability, impr-ove PNS remission rate, it probablely relates to suppressing excret cytokines of MCs.
