Objective To observe the effect of the UI, mucosal inflammation score with HE dyeing microscopy, the expression of serum eHSP72、the content of Hp IgG,T lymphocyte subsets(CD3+、CD4+、 CD8+and CD4+/CD8+)of Hp gastritis in rats with Moxibustion pretreatment, to clarify the immunological mechanisms of protect the gastric mucosa in rats with moxibustion pretreatment,to provide an experimental basis for moxibustion protect the gastric mucosa and moxibustion prevention disease。Methods40healthy SD rats were randomly divided into4groups, namely, blank group (Group A), Hp model group(group B), moxibustion at acupoints groups (group C)and electro-acupuncture acupoints group (group D),10rats in each group, to made gastritis model with Hp Gavage, give Moxibustion and electroacupuncture treatment at seven days before modeling and molding period, once a day, a total of sixteen days,then all rats were sacrificed at twenty-eight days last gavage,to observe UI, Hp rapid urease test, gastric mucosal smear gram’s stain, the inflammation score of gastric mucosa HE staining.to delect the content of serum Hp IgG with enzyme-linked immunosorbent assay, to delect the content of T lymphocyte subsets CD3+CD4+CD8+and CD4+/CD8+with flow cytometry assay.Results1. Compared with group A, the UI were significantly increased in group B(P<0.01); Compared with group B, the UI were significantly decreased in group C and group D(P<0.01);There were no significant difference between group C and D in the UI.2.Rapid urease test positive rate:Compared with the groupA, groupB Quick urea enzyme test positive rate was significantly increased (P<0.01); Compared with groupB, groupC rapid urea the enzyme test positive rate was significantly decreased (P<0.01), groupD quickly ureaenzyme test positive rate decreased slightly, but no significant difference (P>0.05); compared with groupD,group C rapid urease enzyme test positive rate decreased (P<0.05).3.Pathological score of stomach tissues:compared with group A,pathological score was higher in group B(P<0.01); compared with group B, pathological score was lower in group C(P<0.01),but had no significant difference between group B and group D; compared with group D, pathological score was lower in group C(P<0.01).4. Gastric mucosal smear Gram stain examination:groupA gastric Gram stain microscopy did not find a red curved or spiral bacteria(Hp); compared with the groupB, groupC after the gastric mucosa stained microscopic examination shows that a large number ofthe red curved or spiral bacteria (Hp); groupC and groupD the gastric mucosa Gram staining red curved or spiral bacteria (Hp) was significantly reduced compared with the model group.5.Morphological changes of rat gastric tissue:Gastric tissue structure was integrity, is complete, no damage appeared in group A.. In Group B and group D, gastric mucosa was damaged, shedding, lamina propria glands in the shallow layer is damaged, a few inflammatory cell infiltration, gastric mucosal injury obviously; compared with group B and group D, group C rat gastric mucosal damage, shedding more light, less inflammatory cell infiltration.6. Contents of serum Hp IgG:Compared with group A, the expression of Hp IgG in group B were significantly increased(P<0.01). Compared with group B, the expression of Hp IgG in group C were significantly decreased(P<0.01), but in group D were no significantly decreased(P>0.05);Compared with group D, the expression of Hp IgG in group C were significantly decreased(P<0.01).7. Contents of plasmaT lymphocyte (CD3+、CD4+、CD8+) and ratio of CD4+/CD8+:Compared with group A, the content of CD3+、CD4+and the ratio of CD4+/CD8+in group B decreased significantly decreased(P<0.01)., the content of CD8+.increased(P<0.01).Compared with group B, the content of CD3+CD4+and the ratio of CD4+/CD8+in group C increased significantly (P<0.05), but in group D were no significantly increased(P>0.05);Compared with group D, the content of CD3+、CD4+、CD8+have no significantly increased(P>0.05),butthe ratio of CD4+/CD8+in group C increased significantly (P<0.05).Conclusion (1) Moxa Moxibustion can significantly descrease the injury index of gastric mucosa in rats Hp gastritis, reduce gastric mucosa HE staining degree of inflammation integral, prompting Moxa Moxibustion can reduce gastric mucosal injury in rats Hp gastritis..(2) Moxa Moxibustion can significantly increase the content of plasma CD3+、CD4+and the ratio of CD4+/CD8+, descrease the expression of serum Hp IgG, prompt mechanism of Moxa Moxibustion on gastric mucosal protection may enhance the body immunity, resistance to microbial pathogens invasion and removal of Hp has been invaded by Hp, inhibiting the formation of Hp IgG.(3) Moxa Moxibustion and electro-acupuncture group can reduce the UI of gastric mucosa, protect gastric mucosal. But the Moxa Moxibustion group in the enhancement of immunity and reduce the Hp IgG is better than that of electro-acupuncture group.
