Chinese herbal recipe is a major clinical form of Chinese Traditional Medicine (TCM). Research on the active fraction of Chinese herbal recipe helps us understand the material foundation of recipe's pharmacology and explain the essence of multicompoent of Chinese herbal recipe. As one of commonly encountered diseases in clinic of TCM, Pi-deficiency is one of the main syndromes of digestive system diseases and associated with intestinal microflora. In this research, a new molecular evaluating system for"Pi-deficiency"was established by ERIC-PCR method and used for screening the active fraction of three"Pi-deficiency"therapy Chinese herbal recipes. And the active extracted fractions were preliminarily determined."Pi-deficiency"model rats were induced by water extract of Folium sennae and Radix et Rhizoma Rhei respectively. The total DNA of microbial communities in rat fecal samples were amplified using the ERIC primers to generate ERIC-PCR genomic fingerprints. By comparing the ERIC-PCR fingerprints results during healthy and diseased phases, the molecular evaluating system which includes characteristic band (net integral area and abundance), Shannon's index and similarity coefficient was established. Acetic ether fraction, n-buatol fraction and water fraction of three"Pi-deficiency"therapy Chinese herbal recipes: Si-jun-zi-tang, Li-zhong-tang and Bu-zhong-yi-qi-tang were evaluated using the molecular evaluating system. It's the first reported that the intestinal microflora molecular evaluating system was used in evaluating and screening the active fraction of"Pi-deficiency"therapy Chinese herbal recipes.
