Objective:To explore the effects and mechanism of of NaoTai Fang on protection of blood-brain barrier after focal cerebral ischemia in ratsMethods:80SD adult rats were randomly divided into6groups averagely: sham group, large dose group of NTF (27g/kg), medial dose group of NTF (9g/kg), low dose group of NTF (3g/kg) and model group. After corresponding therapy by intragastric administration for3days, rats received the regional cerebral ischemia by the method of middle cerebral artery occlusion (MCAO) with suture. after following3days intragastric administration as previous, Zea Longa ZE neurological defect score was used to analyse the neuroethology of rats; HE staining was used to detect the pathology of brain; TTC method was used to detect the infarct area of the brain; the contents of MMP-9proteins in serum were measured by enzyme-linked immuno sorbent assay(ELISA) method with marketable kit; the expression of MMP-9in brain was measured by immunohistochemical methodResults:The neurological defect score in the large dose group of NTF was significantly decreased than model group (P<0.05); in the large, medial, low dose group of NTF, the ratio infarct area of the brain were significantly decreased than model group (P<0.05), the expressions of MMP-9in serum and brain were decreased than model group (P<0.05) Conclusion:NaoTai Fang can protect the blood-brain barrier after regional cerebral ischemia in rats through the mechanism of decreasing MMP-9expression. |
