Protective Effects Of Hua Yu Ke Sai Fang Pretreatment On Focal Cerebral Ischemia Injury In Rats And Its Mechanisms

Objective:The aims of this study are to investigate the protective effects and its possible mechanisms of Hua Yu Ke Sai Fang pretreatment on the brain after ischemia injury in rats using the model of middle cerebral artery occlusion(MCAO).Methods:75 rats were randomely divided into five groups(15/group):sham-operated group,ischemia group,Nao Xin Tong group(0.48g/kg),high dose Hua Yu Ke Sai Fang pretreatment group(24.4g/kg)and low dose Hua Yu Ke Sai Fang pretreatment group(12.2g/kg).Hua Yu Ke Sai Fang groups were treated by Hua Yu Ke Sai Fang and Nao Xin Tong group were treated by Nao Xin Tong 7 days before MCAO,,2/day; whereas sham-operated group and ischemia group were given NS.6 hours after ischemia,rats were sacrificed by heads cutting and brains removing for the following studies:1)Five rats in each group were used to estimate the volume of brain ischemic damage.Then they were used for determination of ET,NO and P selection;The sections of the optic chiasma brain were stained infarct volume at the coronal level of the rostralin TTC,then the edge of the optic chiasm was calculated by image analysis; 2)Five rats in each group were used for HE stain.At the same time,heat shock protein 70(HSP70)expression were detected by immunohistochemistry;3)Five rats in each group were used for determination of the expression of the mRNA level of heat shock protein 70 by RT-PCR method.Results:Compared with sham-operated group,the neurological deficit and infarction volume were increased significantly after ischemia injury.The percentage of apoptosis and intracellular calcium content were also obviously increased.Noticeable ischemic phenomenon was also seen in HE stain.The expression of ET and P selection were increased significantly,while NO were decreased remarkbaly.The expression of HSP70 and HSP70mRNA were significantly increased.In comparing with ischemia group,Hua Yu Ke Sai Fang and Nao Xin Tong can decrease the expression of ET and P selection,and increase the expression of NO.At the same time,they can decrease the expression of HSP70 and HSP70 mRNA.Conclusion:These results suggest that Hua Yu Ke Sai Fang can attenuate brain ischemia by improving neurological deficit and decreasing infarct volume.So in rats,Hua Yu Ke Sai Fang can protect the brain from ischemia injury,and the neuroprotectionsm echenism may be related to the decrease of ET,P selection and the increase of NO. Inhibition of the expression Of HSP70 mRNA and protein by Hua Yu Ke Sai Fang may be related to its effect of brain protection against cerebral ischemia.
