Objective:To investigate the impact of gastric tumor cell line SGC-7901on the transformationfrom M1to M2,via vitro co-culture of gastric tumor cells SGC-7901with classicactivation of macrophage and alternative activation of macrophages.Methods:We isolated monocytes from peripheral blood by full-automatic magnetic beadscollecting and purification system.GM-CSF and M-CSF were used to induce themonocytes respectively to M1and M2.The Immunohistochemistry was used respectivelyto observe the percentage of CD68, CD16, CD206molecule expression of monocytes,M1and M2. ELISA method was used to detect IL-10and IL-12concentration in supernatant ofmonocytes, M1and M2.Results:CD68molecule was highly expressed on monocytes, M1and M2.CD16molecule wasexpressed more highly on M1, CD206molecule was expressed more highly on M2. Butafter the co-culture with gastric tumor cells SGC-7901, CD206molecule was expressedmore highly on M1then before.The level of IL-12secreted by M1was obviously higherthan that secreted by M2(P<0.05). The level of IL-10secreted by M1was obviouslylower than that secreted by M2(P<0.05). The level of IL-10secreted by M1wasimproved after the co-culture.Conclusion:In vitro culture, gastric tumor cells SGC-7901can advance the transformation fromM1to M2.
