Objective:To identify the phenotype of peritoneal macrophages from different degrees of stomachwall invading of gastric cancer and confirm the relation between the phenotype of peritoneal macrophages and the degree of stomachwall invading of gastric cancer. Methods:The peritoneal washings from patients with different degrees of stomachwall invading of gastric cancer and inflammation were collected, and the peritoneal macrophages were isolated by Ficoll density gradient centrifugation method and cultivated. They were divided into inflammation group, T1-3group and T4group, and every group were10patients. To identify the phenotype of macrophages, IL-10and IL-12were detected by using ELISA method and CD16and CD206were detected by using Flow Cytometry method. The data were collected and analysed by the software SPSS17.0.Results:(1) The results of ELISA showed that the concentrations of IL-10and IL-12were no statistically significant differences among the three groups on36h (FIL-10=1.215, FIL-12=1.767, P>0.05). The concentration of IL-10in T4group was highest (F48h=147.586, F72h=156.365, P<0.05), however their was no statistically significant difference between inflammation group and T1-3group (P>0.05) on48h and72h. The concentration of IL-12in T4group was lowest (F48h=30.707, F72h=143.191, P<0.05), however their was no statistically significant difference between inflammation group and T1-3group (P>0.05) on48h and72h.(2) The concentrations of IL-12in inflammation group and T1-3group were both higher than the concentrations of IL-10(P<0.05), however the concentrations of IL-10in T4group were higher than the concentrations of IL-12(P<0.05) on48h and72h.(3) The results of Flow Cytometry showed that the number of CD16positive macrophage in T4group was least (F=832.148, P<0.05), and their was no statistically significant difference between inflammation group and T1-3group (P>0.05). The number of CD206positive macrophage in T4group was largest,(F=640.346, P<0.05), and their was no statistically significant difference between inflammation group and T1-3group (P>0.05).(4) The number of CD16positive macrophage in inflammation group and T1-3group were larger than the number of CD206positive macrophage (Tinflammation group=34.75, TT1-3group=34.004, P<0.05), however the number of CD206positive macrophage in T4group was larger than the number of CD16positive macrophage (TT4group=31.951, P<0.05).Conclusion:(1) The peritoneal macrophages with gastric cancer patients of invading serosa (classification T4) are mostly CD206+and IL-10+, and the phenotype is mostly M2. The peritoneal macrophages with inflammation patients and gastric cancer patients of no invading serosa (classification T1-3) are mostly CD16+and IL-12+, and the phenotype is mostly M1.(2) The number of CD206+, IL-10+macrophage (M2macrophage) increases because stomachwall is permeated by gastric cancer.
