Objective(1)Study the anti-aging effect of perilla oil on D-galactose induced aging mod el rats by the observation of physical ability, the ability of learning memory, antioxidant and lipid peroxidation index, and research the anti-aging mechanism preliminarily.(2)Study the mechanism of promoting learning and memory ability of perilla oil by observing the effect on rat hippocampal expression of acid-sensing ion channels, and explain the of it’s anti aging mechanism.Method(1) A total of40male SD rats of clean grade,1month old, were randomly divided into5groups. Negative control group, aging-model group, small dose perilla oil group (0.815ml/kg), middle-dose perilla oil group (2.445ml/kg) large-dose perilla oil group (7.337ml/kg). Except for the negative control group were injected with saline, the remaining rats in each group in the subcutaneous injection of D galactose (125mg/kg). After8weeks’feeding, weight loading swimming test and Morris water maze experiment was carried out, then determine the content of superoxide dismutase (SOD) and malondialdehyde (MDA) and nitric oxide synthase(NOS) and nitric oxide(NO).(2)A total of32male SD rats,1month old, were randomly divided into4groups, each group consisting of8rats. Small dose perilla oil group0.815ml/kg), middle-dose perilla oil group (2.445ml/kg), large-dose perilla oil group (7.337ml/kg) and a negative control group. Perilla oil was fed by orogastric intubation within30days, then step-down task was carried out, ASICs1a, ASIC3and ASIC4mRNA expression was observed in hippocampus of rats by RT-PCR method.Result:(1) Compare with aging model group, the time of swimming in small-dose perilla oil treatment group, middle-dose perilla oil treatment group and large-dose perilla oil treatment group were longer(P<0.05); the escape latency of small-dose perilla oil treatment group, middle-dose perilla oil treatment group and large-dose perilla oil treatment group were less than model group (P<0.05); the effective time ratio in small-dose perilla oil treatment group, middle-dose perilla oil treatment group and large-dose perilla oil treatment group was higher than model group P<0.05), The time of going through the platform in small-dose perilla oil treatment group had no difference, In middle-dose perilla oil treatment group and large-dose perilla oil treatment group, the effective time ratio and the time of going through the platform were remarkably higher than model group (P<0.05); compared with model group, In all groups the SOD activity was remarkably higher than model group(2) Compared with control group, the expression of ASICla, ASIC3and ASIC4mRNA was improved by each dose of Perilla oil group。Conclusion(1)Successfully copy the D-galactose subacute aging model; Perilla oil can improve learning memory ability of D-galactose subacute aging model, increasing SOD activity and decreasing MDA content, indicating that Perilla oil have anti-aging effect. Perilla oil anti-aging efficiency was operated by reducing the blood free radicals.(2)Perilla oil can improve ASICla, ASIC3and ASIC4expression, the expression first increase and then decrease as Perilla Oil dose increased; The promoting learning and memory ability of perilla oil related to ASICla.
