ObjectiveTo observe related cytokine secretion and protein expression in colorectalcancer cell line SW620which non-contact co-cultured with M2typemacrophage, in order to investigate the effects of epithelial-mesenchymaltransition on SW620by M2type macrophage.MethodsInduce human leukemic cell line THP-1into M2type macrophage andconfirmed by flow cytometry. Non-contact co-cultured M2type macrophagewith SW620as study group, SW620cell alone as control group. Use ELISAassay to detect TNF-α and TGF-β cytokines in cultural supernates in every24hs for3days respectively. Use western blot assay to measure E-Cadherinand Vimentin expression on SW620in the same time pattern.ResultsResults from ELISA assay show TNF-α and TGF-β secretion in studygroup is higher than control group and increased with incubation time.(P<0.05)Results from western blot assay show E-cadherin expression in studygroup is decreased while Vimentin expression is increased with incubationtime.(P<0.05)ConclusionM2type Macrophage can promote epithelial-mesenchymal transition onSW620.
