Background and Purpose: Lung cancer is the world’s highest incidence of cancer,1.2million ofthe annual increase of patients. Lung cancer mortality is highest in China’s large andmedium-sized cities. Non-small cell lung cancer (NSCLC) accounts for more than80%of lungcancer. Although the clinical treatment has made significant progress, the cure rate has been stillabout10%. The pathogenesis of lung cancer is unknown and the effective methods for earlydiagnosis are seldom,have always troubled the clinician. Intermediary subunit Med19in thehomologous gene of human lung cancer metastasis is related to gene (LCMR1,). Med19genelocated at11q12.1, is an important part of the RNA polymerase Ⅱ general transcription unit, itsmajor extracellular roles include promoting transcriptional activation, enhancing basaltranscription, promoteing CTD (carboxyl terminus domain of the polymerase large subunitCarboxy the terminal domain) phosphorylation, directly and indirectly being involved inregulating cell growth, differentiation and apoptosis. In recent years, studies have found increasedexpression of the Med19genes in breast, liver, stomach, and a variety of human tumors. A largenumber of studies have shown that Med19is involved in a variety of tumor development process,but current research focuses on the in vitro stage, less clinical research, in particular, is still blankin lung cancer. By detecting different Med19expressions in non-small cell lung cancer andadjacent tissues and its relationship with Ki-67, we try to explore the role of the Med19protein inoccurrence and development of lung cancer, and application prospects in lung cancer diagnosis,and treatment.Methods: The study randomly selected primary lung adenocarcinoma and lung squamous cellcarcinoma patients of radical or palliative resection with complete clinical and pathological data of Cancer Prevention and Research Institute of Shandong Province, from February1,2010to July31,2011, a total of56cases. For their lung cancer tissue samples and cancer adjacent tissuesamples as the control group, Med19mRNA content is measured by real-time PCR andMed19and Ki-67protein expression was detected by immunohistochemical assay.Meanwhile therelationship between Med19and Ki-67protein expression and a variety of clinical pathologicalfeatures including age, gender, pathological type, histological grade, lymph node metastasis,clinical stage (according to the relationship between the International Union Against Cancer2003TNM installments)was analyzed by statistical methods. The Med19protein expression innon-small cell lung cancer clinical significance and relationships with Ki-67was explored. Andan effort was made to provide the basis for the Med19becoming a potential target for treatment oflung cancer.Results: Med19m RNA expression in NSCLC lung cancer was significantly higher than thepaired adjacent normal tissues.Med19protein positive expression rate in cancer-adjacent tissues isonly1.8%, compared with adjacent tissues, Med19protein expression intensity in cancer tissuesincreased significantly, which is mainly the nucleus-coloring, positive expression rate is80.4%.Cancer tissue Med19positive rate is significantly higher than the cancer adjacent tissues (P <0.05),suggesting that Med19protein related to the occurrence of lung cancer. The med19proteinexpression in NSCLC patients is related to clinical stage and lymph node metastasis (P <0.05),but not with other clinicopathological parameters such as age, gender, histological type andhistological grade (P>0.05), relationship between Med19protein expression and ki-67proteinexpression in cancer tissues is positively correlated.Conclusion: Med19is closely related to abnormal mitotic of lung cancer cells. Med19proteinenhanced expression prompts malignant degree and increased transfer activity of lung cancer. TheMed19involved in different stages of development of lung cancer, is expected to become aimportant basis of clinical diagnosis and staging of lung cancer, and can be used as a new tumortherapeutic targets.
