Mulberry leaf is dry leaf of Morus alba L, and it is for medical and alimental use. Study showed that mulberry leaf has many Pharmacological activity such as lowering blood sugar level, reducing blood pressure, reducing blood fat, anti-tumor, antioxidation, etc, Among these functions, the main effective component are flavone, alkaloid and polyose ingredient. Thus, it has very important academic significance and practical value, for developing new medicine of diabetes mellitus and developing related mulberry leaf health products, to extract, isolate, and purify mulberry leaves to get DNJ, flavone, polysaccharide and to establish a method to measure its content. The study result of this thesis is as the following:First, all polysaccharides TPM of mulberry leaf can restrain blood sugar rises after meal; all alkaloids, especially DNJ(1-deoxynojirimycin) is a nature a-glycosidase inhibitor; general flavone such as rutin, quercetin, and quercitrin can improve blood vessel endothelium and has the function of lowering blood sugar and eliminating oxygen radicals. Total polysaccharides, general flavones and total alkaloids in the mulberry leaf have remarkable effect of lowering blood sugar. The author, for the first time, separated the above active various kinds of ingredients of lowering sugar, and this can save medicine resources greatly and increase utilization of mulberry leaves. Though orthogonal test comparison, the best technology of separating glucose-lowering active ingredient is to adopt acid extraction first and alkali extraction second, which is ultrasound aided solvent method. The best synchronous extraction technology of mulberry alkaloid and polysaccharide is that the time is35minutes, with temperature of60℃and the PH value is1.0, the solid-liquid ratio is1:12(m/V), the extraction time is3; the best condition of extraction of flavone from residue is:that the time is35minutes, with temperature of40℃and the PH value is12.0, the solid-liquid ratio is1:10(m/V). Besides, by means of spectrophotometry, the content of total polysaccharides, general flavones and total alkaloids are measured, and scientific proof is provided to reasonable utilization of mulberry leaf resources.Second, in the thesis, for the first time, microscopy method is introduced to compare form differences of before and after vacuum broke, compare content differences of effective ingredients of glucose-lowering, which are1-deoxynojirimycin, general flavore and polysaccharide, and study the influence which vacuum broke technology has on glucose-lowering ingredient extraction and isolation technology of mulberry leaf.Finally, the thesis compares the inhibitive function to pathogenic microorganism among various kinds of mulberry leaf extracts...
