Diabetes is a chronic which is harmful to people's health. It is the third factor leading human to death, inferior to cardiovascular disease and tumor. Mulberry leaves Which have the particular pharmacological activities and high efficiency of lowing blood glucose without obvious toxicity and side effect become the hot of studying and inventing new antidiabetic drugs. Shart first reported that the product of hot water extract from mulberry leaves have pharmacological activities of lowing blood glucose and people in China also proved that. People speculated that the effective part were relation to its alkaloids or flavonoids,while polysaccharides'pharmacological activities of lowing blood glucose was also reported. Because of lack of true effective part, so it is necessary to find out. Therefore, , withdrawing the alkaloids,polysaccharides,flavonoids extracted with hot water from mulberry leaves to combine to compare its pharmacological function to find out the most effectively respectively part is very meaningful. The contents of this article are as follows: (1) The alkaloids were extracted from mulberry leaves with boil water , collected by cation exchanger column chromatography(H+form,001×7), separated and purified by the 0.5moL·L-1NH3·H2O .The elute was extracted by n-butanol, A (alkaloids) were obtained by concentrating the n-butanol extracted. The content of alkaloids was determinated as 27.1% with the weight method. (2) The crude polysaccharides were obtained from the extraction of boil water from mulberry leaves through polymer resin D101 absorption,ethanol precipita tion .The crude polysaccharides were purified by means of carbon decolored ,Sevag deprotained . Thus B(polysaccharides) was obtained. The content of polysaccharide was measured by anthrin-H2SO4 and the result is 71.8%. (3) The flavonoids were extracted from the leaves of Mulberry by boiling water, collected by polymer resin D101 absorption , separated and purified by 80% alcohol. Then we refined the flavonoids. The content of flavonoids was determinated as 44.2% by HPLC. (4) Two kinds of flavone aglycones were obtained from hydrolyzed refined flavone by water , through silica gel chromatography separation. On the basis of chemical and spectroscopic evidence, their structure were verified as quercetin and kaempferol. (5) Four kinds of flavone glycosides were obtained from C through silica gelchromatography separation ,TLC. On the basis of chemical and spectroscopic evidence, two compounds'structure were verified as kaempferol -3-O -β-D -glucoside quercetin-3-O -β-D -glucoside。(6) The extract of A(alkaloids),B(polysaccharides ),C(flavnoids),C'l,C'2,C'3 (C'l,C'2,C'3 were compounds of flavnoids) were carried on pharmacology's experiment of lowing blood glucose 。The result shows that B can decline the blood glucose obviously and improve the diabetes'symptom 。The other compounds have little effect。...
