The Effect Of Litchi Flavonoids On Proliferation Of Rat Hepatic Stellate Cells And The Expression Of Protein TGF-β₁and α-SMA

Objective:The aim of this study was to investigate the molecular mechanism and the effect of growth inhibition on Litchi flavonoids (TFL) to rat hepatic stellate cells (HSC-T6).Methods:MTT assay was used to test Rat hepatic stellate cells (HSC-T6) growth inhibition, and the cell apoptosis was observed after stained with Acridine orange; Cell cycle was detected by Flow cytometry.Results:A variety concentration of Litchi flavonoids (TFL)can inhibit growth activity on rat hepatic stellate cells (HSC-T6) in different time (P<0.05),and the half inhibitory concentration was101.98mg/mL; In the groups treated by TFL, nuclear enrichment, nuclear breakdown and dense-stained bright green apoptotic bodies were found. Application of flow cytometry and drug treatment control group the distribution of cell cycle, the test results show that, Control cells G1phase cells as47.68±2.92%, G2phase cells as19.58±2.93%, S phase cells as32.75±2.09%; drug group by Litchi flavonoids (TFL) treatment, G1phase cells increased, while the G2phase cells were significantly reduced, the cells were obviously arrested at G2phase (P<0.05). Western Blot detection of cells in the application of TGF-β1and a-SMA protein expression showed that, With the increase of drug concentration, TGF-β1and a-SMA protein expression was decreased, compared with the control group,100mg/ml and120mg/ml of drug concentrations of TGF-β1and a-SMA protein expression was significantly reduced (P<0.05)Conclusions:Litchi flavonoids (TFL) can inhibit proliferation, promote differentiation and induce apoptosis on rat hepatic stellate cells (HSC-T6).
