Obejectives:The inhibitory effect of total flavonoids from litchi nucleus on the proliferation of rat hepatic stellate cells,and the possible mechanism of anti-hepatic fibrosis induced by total flavonoids from litchi nucleus from TGF-?1/Smad pathway and PPAR-r/c-Ski pathway.Methods:In vitro culture of HSC-T6 cells,Divided into blank control group,TFL concentration 20ug/ml group,40ug/ml group,80ug/ml group,160ug/ml group,320ug/ml group,After 48 hours of intervention,CCK-8 method was used to detect the proliferation of each group of cells;then set to blank group,colchicine control group,TFL low dose group,TFL medium dose group,TFL high dose group ELISA method to detect each group of cells The contents of PPAR-r and c-Ski were detected in the supernatant;the expression of TGF-?1 and Smad3/4 mRNA in each group was detected by qPCR.Results:The results of CCK-8 showed that the inhibition rate of drug proliferation increased with the increase of TFL concentration;The results of ELISA showed that the levels of PPAR-r and c-Ski in the supernatant of each group were significantly higher than those in the blank group(P<0.01),and the highest in the TFL high-dose group(P<0.01);Compared with the colchicine control group,the levels of PPAR-r and c-Ski in the supernatant of the TFL high-dose group were higher than those in the colchicine control group(P<0.01),and the cell supernatant in the TFL medium-dose group.The PPAR-r content was relatively higher than that of the colchicine control group(P<0.05).The cell supernatant c-Ski content in the TFL medium dosegroup was higher than that in the colchicine control group(P<0.01),and the cell supernatant supernatant PPAR-r content in the TFL low dose group was lower than that in the colchicine control group(P <0.01),the cell supernatant c-Ski content in the TFL low-dose group was relatively higher than that in the colchicine control group(P<0.05);qPCR results showed that TGF-?1,Smad3 in each group of cells compared with the blank group The expression of /4 mRNA was significantly decreased(P<0.01),and reached the lowest in the high dose group(P<0.01).Compared with the colchicine control group,the expression of TGF-?1 and Smad3/4 mRNA in the high-dose TFL group was significantly lower than that in the colchicine control group(P<0.01),and in the TFL medium-dose group.The expression of Smad3 mRNA was significantly lower than that of the colchicine control group(P<0.01).The expression of TGF-?1 and Smad4 mRNA in the TFL medium dose group was lower than that of the colchicine control group(P<0.05).The expression of TGF-?1 mRNA in the low-dose TFL group was lower than that in the colchicine control group(P<0.05).The expression of Smad3/4 mRNA in the low-dose TFL group was higher than that in the colchicine control group.Group(P < 0.05).Conclusion:The total flavonoids from litchi nucleus can inhibit the proliferation of HSC-T6 cells,Reduce extracellular matrix secretion,This mechanism may increase the content of PPAR-r and c-Ski in the supernatant of HSC-T6 cells,Inhibition of TGF-?1,Smad3/4 mRNA expression.
