The product of P53 gene is a kind of phosphoprotein, which is located in cytoblastand is cell cycle associated protein, P53 protein can bind DNA sequence specifically andactivate some genes, which participate in regulating cell cyle and suppress abnormal cellinto S cycle and induce cell apoptosis and inhibit expression of some genes that arerelevant to cell malignant proliferation and suppress angiogenesis and inhibit metastasisof tumor cells et al. P53 protein has wild type and mutant type, Wild type p53 proteinexpresses lower in all normal cells, while mutant type P53 protein expresses higher intumor cells. MDM2(murine double minute 2) protein is a kind of DNA binding protein,repressing wild type P53 to inhibit cell transformation and possessing transcription andregulation, The biological role of MDM2 can strengthen cell longevity, extending celllife span, facilitating cell proliferation and tumor growth. The research discovered thatthe function of MDM2 is syntrophic with P53, Activated P53 leads to excessiveexpression, while expression of MDM2 suppresses wild type P53 stimulating expressionof target gene. MDM2 expresses or proliferates in malignant tumors such as soft-tissuesarcoma,bladder carcinoma and so on. In recent years, studies indicate thatPTEN(phosphatase and tension homology on chromosome ten) is tumor suppressorgene,which possesses activity of lipid phosphatase first at present, PTEN protein ismuitifunctional protein and is able to inactivate its substrate (PIP3), which isdephosphorylated, Growth inhibitory activity of PTEN is relevant to Glcycle kinase activity lower of cell growth cyle. The research discovers that PTEN gene mutates inhuman many kinds of tumors and hereditary tumor sensitive syndrome and so on, Theexpression of PTEN is relevant to prognosis of tumor patients. At present the researchof PTEN has been original hot spots in tumorigenesis, metastasis and prognosis and soon.Uterine cervix cancer is the most frequent female malignang tumor and has halfnumberabove in malignant tumors of female reproductive system and threaten femalehealth and life seriously. Because uterine cervix cancer has longer lesion precancerous,the mechanisms of occurrence,development of discussing uterine cervix cancer havebeen one of hot spot of researchs. To this date the expression of MDM2,PTEN和P53protein and its relationship havenot relevant documents reports. In order to discuss themechanisms of uterine cervix cancer, search marker, which can utend early diagnosis,early treatment of uterine cervix cancer and lower morbidity and mortality of uterinecervix cancer. Our research adopts immunohistochemistry method, observing theexpressions of MDM2,PTEN和P53 protein in uterine cervix carcinoma tissues of 50cases,adjacent non-atypical hyperplasia tissues of 30 cases and normal mucosa tissuesof 50 cases Our research explores furtherly the mechanisms of occurrence,development of uterine cervix carcinoma and provide rationale for early diagnosis,early treatment of uterine cervix cancer.Methods1. Using the technology of immunohistochemistry SP to detect the expression ofMDM2,PTEN and P53 protein in the 50 cases of squamous cell carcinoma of uterinecervix, 30 cases of atypical hyperplasia tissues of tumor-adjacent and 50 cases of normalmucosa of uterine cervix respectively.2. Statistical analysis: All the dates were analyzed by SPSS 11.0 statistical package,the count information calculated the positive rate, The comparison of positive ratesuses the Chi-square; The relation of two variable are analyzed by the spearman levelcorrelation analysis. The level of significant difference is a=0.05.Results1. Immunohistochemical results indicated: the positive expression rate of MDM2 protein in normal mucosa of uterine cervix is 0%(0/50); The positive expression rate ofMDM2 protein in atypical non-hyperplasia tissues is 33.3% (10/30); The positiveexpression rate of MDM2 protein in the carcinoma tissues is 64%(32/50), the positiveexpression rate of MDM2 protein in the carcinoma tissues is compared with that in thepropotional adjacent tissues and normal mucosa tissues from each other, There isstastital significance(P<0.01).2. Immunohistochemical results indicated: the positive expression rate of P53protein in normal mucosa of uterine cervix is 0%(0/50); The positive expression rate ofP53 protein in atypical non-hyperplasia tissues is 30% (9/30); The positive expressionrate of P53 protein in the carcinoma tissues is 62% (31/50), the positive expression rate ofP53 protein in the carcinoma tissues is compared with that in the propotionai adjacenttissues and normal mucosa tissues from each other, There is stastital significance(P<0.01).3. Immunohistochemical results indicated: the positive expression rate of PTENprotein in the carcinoma tissues,atypical non-hyperplasia tissues and normal mucosaof uterine cervix is 40(20/50),70% (21/30) and 100% (50/50) respectively, the positiveexpression rate of PTEN protein in the carcinoma tissues is compared with that in thepropotional adjacent tissues and normal mucosa tissues from each other, There isstastital significance(P<0.01).4. The relativity of the expression of MDM2,P53 and PTENwas analysized bystatistics, Results showed that the both former is also negative correlation with later(P<0.01), There is a positive correlation between former both(P<0.01).Conclusions1. The positive rates of the expression of MDM2 protein in squamous carcinomatissues of uterine cervix,atypical hyperplasia tissues of the tumor-adjacent and normalmucosa tissues were high in turn, Therefore the comparison of the expression of MDM2protein among carcinoma tissues and adjacent atypical hyperplasia tissues,normalmucosa tissues has statistical signicance. It indicaed that MDM2 participated inoccurrence and development of squamous carcinoma of uterine cervix.2. The positive rates of the expression of P53 protein in squamous carcinoma tissues of uterine cervix,atypical hyperplasia tissues of the tumor-adjacent and normal mucosatissues were high in turn, Therefore the comparison of the expression of P53 proteinamong carcinoma tissues and adjacent atypical hyperplasia tissues,normal mucosatissues has statistical signicance. It indicaed that P53 participated in occurrence anddevelopment of squamous carcinoma of uterine cervix.3. The positive rates of the expression of PTEN protein in squamous carcinomatissues of uterine cervix,atypical hyperplasia tissues of the tumor-adjacent and normalmucosa tissues were low in turn, Therefore the comparison of the expression of PTENprotein among carcinoma tissues and adjacent atypical hyperplasia tissues,normalmucosa tissues has statistical signicance. It indicaed that PTEN participated inoccurrence and development of squamous carcinoma of uterine cervix.4. The relativity of the expression of MDM2,P53 and PTEN protein was analysizedby statistics, Results showed relative and they played synergistic effect during theprocess of occurrence and development of squamous carcinoma of uterine cervix.
