Mayi rhinitis spray is a compound formulation which consists of Ephedrae Herba, Magnoliae Flos, Asari Radix et Rhizoma, Phellodendri Cortex, Syntheticum Borneolum, and Camphor. The preparation is produced by non prescription has become rhinitis water, and then improved and made of pure Chinese herbal compound. According to clinical use for many years, it has good effect on acute rhinitis, chronic rhinitis and allergic rhinitis and sinusitis and so on. In order to further the development and utilization of this traditional Chinese medicine preparation, the preparation of this study were pre-clinical research. The study bases on the Guidance of Provision for New Drug Approval and combines with the advanced formulations of traditional Chinese medicine, includes optimization of preparation process, quality standards and pharmacodynamic study. The ultimate goal of this study is to develop new drugs.Extraction Study: The optimal preparating conditions were screened by orthogonal design L9(4~3). Orthogonal test was designed with the content of ephedrine hydrochloride as index. The results showed that the optimized technology is repeatable and feasible. Establish quality standard: The contents of ephedrine hydrochloride and berberine hydrochloride in the Mayi Rhinitis Spray were determined by HPLC, and the qualitative analysis for Ephedrae Herba, Magnoliae Flos and Phellodendri Cortex were performed using TLC. The qualitative analysis for the volatile components of Mayi Rhinitis Spray was performed using GC-MS. Experiments established the requirements the content range of ephedrine hydrochloride and berberine hydrochloride, and the qualitative identification of control.Finally established for the quality control standards of Mayi Rhinitis Spray.Pharmacodynamic study: The mothods of anti-inflammatory effects were common test method treatment for drug rhinitis. Animal model of acute and subacute inflammation were used to test and verify the anti-inflammatory effects of Mayi Rhinitis Spray. To explore into the pharmacodynamic effect of Mayi rhinitis spray on guinea pig models with allergic rhinitis (AR). The results showed that Mayi rhinitis spray posses anti-inflammatory effects. And there was significant curative effect in guinea pig with AR by treatment of Mayi Rhinitis Spray. Compared with the same type of medicine, Mayi Rhinitis Spray has a better therapeutic effect on experimental allergic rhinitis.
