| Objective Explore the molecular biological mechanism that the changing of local immune environment after peripheral nerve damage,"blood-neural barrier" destruction, which affect the repairing of peripheral nerve damage. To provide the basement formulated for more specific and effective clinical treatments.Methods:44Wistar rats were randomly divided into the experimental group (n=20), control group (n=20) and normal group (n=4). In the experimental group, the right sciatic nerve were cut off, and anastomosised the nerve end to end; in the control group just free out the sciatic nerve only. Normal group did not do any treatment. In one, two and four weeks after the operation, take the sciatic nerve, H E dyeing and the SABC immunohistochemical method were used to show the expression of myelin alkaline protein (MBP), axon growth inhibitory factor (NOGO-a), peptides transformation growth factor (TGF-beta). The dyeing results was statistical analizied.Result The myelin alkaline protein (MBP) were significantly expression in all of the specimens, but experimental group was the most intensely. The experimental group in different time points in sciatic nerve MBP positive express strength1week>2weeks>4weeks. Despite there is a difference in the expression strength of the MBP, but difference have no significance after statistics analysis (Q inspection). TGF-beta can be weakly positive expressed in normal group organization cells. In the control group and the experimental group, TGF-β, are obviously expression, but over time gradually reduced. The statistics analysis (Q inspection) show that there is statistical significance within the group comparing. There are almost no expression of NOGO-a in normal group and the control group.In experimental group the sciatic nerve NOGO-a positive in expression:1 week<2weeks>4weeks.
