The Effect Of Xiaogutong Decoction On The Expression Of Dickkopf-1and IL-1β In Rat Osteoarthritis

ObjectiveTo investigate the effect of Wnt signaling pathway inhibitor Dickkopf-1in different pathological periods of rat osteoarthritis(OA), respectively.MethodsEstablish the model of rat OA by using the method of improved injecting papain into the cavity of rat knee joints. The cartilage was assessed by Mankin’s. DKK-1mRNA expression checked by Real Time-PCR, The expression of DKK-1in protein in synovium and articular cartilage was detected by immunohistochemistry at different groups. Also the serum DKK-1was assessed by ELISA.ResultsCompared with the model group, the DKK-1mRNA and protein level were increased in OA at different pathological periods(p<0.05).ConclusionThe expression of DKK-1was increased with different pathological periods. DKK-1may act as a biological symbol of OA to assess the severity of the disease. Compared with the model group,the DKK-1m RNA andIL-1βmRNA and protein level were decreased in synovium and in articular cartilagein Xiaogutong treatment group and diclofenac group.There is no statistic differencebetween the Xiaogutong Decoction treatment group and diclofenac group.ConclusionXiaogutong Decoction can inhibit theexpression of DKK-1and IL-1β;Xiaogutong Decoction can ease the process of OA;Xiaogutong Decoction maybeease the OA progress by inhibitting the expression of DKK-1and IL-1β.
