Background:Psoriasis is a common chronic, inflammatory, proliferative skin disease.The characters of psoriasis include hyperplasis and altered differentiation of epidermis,infiltration of leukocytes into both the dermis and epidermis, and dilation and growth of bloodvessels. The etiology and pathogenesis of psoriasis is still not fully clear now. At present,most scholars consider psoriasis is an abnormal immune inflammatory disease which isrelated to immune, inflammation, cell proliferation, apoptosis, and micovacular abnormalities.The WHO defines psoriasis as one of the top ten chronic diseases. Incidence rate amongnatural population is0.1-3%, China annual incidence rate is0.123%, and the incidence in ourcountry is increasing now。There are more than three million patients in China. As it’s highincidence, long duration, easy to relapse (especially for young adults), the patient’s physicaland mental health can be influenced hardly. To research pro-inflammatory factors and thegrowth factors is helpful for revealing the pathogenesis of psoriasis and can instruct theclinical treatment. The related pro-inflammatory factors and growth factors gets more andmore attention. MIF is differentiationed by T lymphocyte, as a complex biological activity ofpro-inflammatory cytokines, in vitro the anti-MIF antibodies inhibited T cell proliferation andthe IL-2formation, in vivo activated T cells inhibited antigen activation and antibodyproduction. COX-2is an important enzyme in prostaglandin biosynthesis process, as the fastlimit enzyme which can catalyze acid into the prostaglandin. Functional, COX-2belongs tothe inos, resting normally, but can be expressed in cytokines, hormone, carcinogens and soon.Many kinds of inducing factors would stimulate it’s rapid expression, so that it canparticipate in multiple pathphysiological processes. VEGF is a vascular endothelial cellspecific, not only can promote vascular endothelial cell proliferation, but also stronglyeffectively increase capillary permeability. At present, the relation between pathologic processof VEGF and psoriasis get more attention. Objective To investigate the relationshipbetween macrophage migration inhibitory factor (MIF), cyclooxygenase-2(COX-2) andvascular endothelia growth factor (VEGF) and psoriasis, and their effect and relation inpsoriasis will be explained. To provide a new method for the treatment of psoriasis. Methods The biopsies were taken from skin lesions in30patients with psoriasis vulgarisand the skin of10normal controls, the expression and distribution of MIF、COX-2and VEGFproteins were detected by immunohistochemical method. The results were analyzed by T testand Spearman rank correlation test of SPSS13.0statistic software.Results1.MIF stainingresults: MIF staining markedly presented in cytoplasm of epidermal keratinocytes and dermallayer inflammatory cells and vascular endothelial cells in psoriasis vulgaris, but expressedweakly in basal cell layer and prickle cell layer of health control. The level of MIF protein is(151.04±6.38), which is significantly higher than that in normal controls (180.05±6.00)(P<0.01).2.COX-2staining results: COX-2staining markedly presented in prickle cell layerand basal cell layer and dermal layer inflammatory cells and vascular endothelial cells inpsoriasis vulgaris, but expressed weakly in basal cell layer and prickle cell layer of healthcontrol. The level of COX-2protein is (140.97±11.52), which is significantly higher than thatin normal controls (199.07±7.34)(P<0.01).3. VEGF staining results: VEGF stainingmarkedly expressed in cytoplasm of epidermal keratinocytes and dermal microvascularendothelial cells in the nipple and dermal layer inflammatory cells. VEGF protein is weaklyexpression in normal controls. The level of VEGF protein is (156.88±6.79), which issignificantly higher than that in normal controls (187.34±4.81)(P<0.01).4. There issignificant positive correlation between the expression of MIF and VEGF(r=0.755,P<0.01).5. There is significant positive correlation between the expression of COX-2andVEGF(r=0.691,P<0.01).6. There is significant positive correlation between the expression ofMIF and COX-2(r=0.681,P<0.01).Conclusion1.MIF was over expressed in psoriasislesions, which suggested that it may be involved in the infiltration of chronic inflammatorycells.2.COX-2was over expressed in psoriasis lesions suggested that COX-2may play animportant role in the occurrence and development of psoriasis as a pro-inflammatory factor.3.VEGF was over expressed in psoriasis lesions suggested that may be involved in theformation of new blood vessels.4.The expression of MIF、COX-2and VEGF in the psoriasislesions are positively correlated with each other which shows that they have the direct orindirect relationships in the development of psoriasis lesions.
