Objective:1.To detect the genetic diversity of Capparis spinosa L. byISSR(inter-simple sequence repeat) molecular markers;2. To develop a new preparation ofChinese Medicine which is safety and effective on pedo-diarrhea, based on the study aboutthe gel ointment of Capparis spinosa L. Methods:1. Selected the sutiable andreproducible primers from100primers and optimized the reaction system of ISSR fittingfor genetic diversity by single factor test design, after the ISSR-PCR amplification,POPGENE and MEGE sofeware were used for genetic diversity anlysis.2. Single factorexperiments was tested and used the content of rutin and alcohol extracts of Capparisspinosa L. as the index to optimize the extraction process; filtered matrix and optimizedthe preparation process for gel ointment, used peeling strength and physical properties asthe indicators; took the quality control according to the Chinese Pharmacopoeia; screenedoptimal transdermal transdermal penetration enhancers by the transdermal experimentsand rutin content as an indicator. Results:1. The ten primers on an average produced94bands across100samples from ten populations of ten regions, of which93werepolymorphic, accounting for98.94%, at the population level, effective al1eles(Ne) were1.4883,Nei’s gene diversity(H) was0.2874, Shannon’s information index(I) was0.4379,the percentage of polymorphic loci was98.94, gene flow among populations(Nm) was0.7093, the coefficient of gene differentiation was Gst=0.4134; based on Nei’s genedier,cluster analysis by UPGMA showed that all populations could be divided into two largegroups.2. The best extraction technology for Capparis spinosa L. was extracted by90%ethanol(Capparis spinosa L.(m):ethanol(v)=1:8)three times,each time1hour; the bestmatrix ratio of gel paste of Capparis spinosa L. was K-30: CMC-Na: glycerol: NP-700:Ganhydroxylaluminum: Tween80=2:0.2:15:3:0.2:1;20mg·g-1Azone enhanced thecumulative permeation quantity better than other penetration enhancers and it,s enhancerratio was1.2575(P<0.05),permeation rate of rutin was5.4457μg·cm-2·h-1. Conclusion:1. The research proved that ISSR molecular markers can reflect the genetic differences and genetic diversity among all Capparis spinosa L. samples.2. The gel ointment ofCapparis spinosa L. provided a theoretical foundation and practical experience for thedevelopment of new formulations of traditional Chinese medicine.
