| Objectives: analyse Expression and mutation of Arid1a in gastric cancerMethods: Collected166cases of gastric cancer specimens which preoperative diagnosis andpostoperative diagnosis were gastric cancer by Affiliated Union Hospital of Tongji Medical College,Department of Pathology among2006to2010.And collected154cases of normal gastric mucosa next tothe cancers.The166cases of gastric cancer patients did not undergo radiotherapy and chemotherapy andfollowed up7months to5years.used immunohistochemical staining method to detect Arid1a protein ingastric carcinoma. Extracted RNA from the specimens,used real time polymerase chain reaction methodto measure mRNA expression in various tissues. Extraction DNA from the specimens and detectedArid1a gene mutation in gastric cancer.Results: Immunohistochemistry test results indicated that in the gastric cancer adjacent normal tissuethe abense of Arid1a expression rate was7.80%, lowerer than24.10%of gastric cancer, the differencewas statistically significant (P <0.01), the abense of Arid1a expression had correlation with tumordifferentiation degree(P=0.0169), lymph node metastasis (P=0.0345), depth of tumor invasion (P<0.0001) and pTNM stage (P <0.0001); real-time quantitative polymerase chain reaction results showedthat in the gastric cancer adjacent normal tissue Arid1a mRNA relative expression was higher than ingastric carcinoma; the survival analysis showed the abense of Arid1a expression patients that mediansurvival time was lower than the positive expression of Arid1a patients (P=0.036). Randomly selected50cases of gastric cancer specimens,6cases of gastric carcinoma were detected mutation; the mutationrate is12%. Conclusion: The absence of Arid1a expression was low in normal tissues,and the absence of Arid1aexpression was high in gastric cancer tissues.the lower degree of tumor differentiation, lymph nodemetastasis, the deeper depth of tumor invasion and the later of TNM stage, the lower of Arid1aexpression, and found that the absence of Arid1a expression was higher, And found that ARID1A lackof expression suggested adverse prognosis.Therefore, Arid1a as a tumor suppressor gene involved in theoccurrence and development of gastric cancer.Randomly selected50cases of gastric cancer specimens,6cases of gastric carcinoma were detected mutation, the mutation rate was12%.which was2%higherthan the foreign literature reported, that may have correlation with race and sequencing methods as wellas the probability of randomly selected specimens. In addition, the different between Arid1a mutationrates and the absence of Arid1a expression suggested that there may be other ways to cause the absenceof Arid1a expression, such as methylation. |
