Sesamin is a kind of active ingredients in sesame seeds. Recent studies dem-onstrated that sesamin has extensive biological activities, such as loweringblood-lipid, anti-oxidation activity and modulating the lipid metabolism etc. it hasstrongly linked with liver damage and liver lipid peroxidation which caused bynonalcoholic fatty liver disease. This paper used mice were fed on high-fat diet for8weeks to make up the non-alcoholic fatty liver model,studying from lipid levelsand free fatty acid composition, oxygen radical metabolism, hepatocyte damageaspects and the gene expression changed by key enzyme of the lipid metabolism.Research the preventive and curative effects and mechanism of sesamin on nonal-coholic fatty liver disease.The results show that:1. The NAFLD mice model was establish with high fatdiet. After8weeks, the model group compared with normal one obviously was de-creased the serum T-CHO and TG, increased the serum HDL-C and hepatic steato-sis.2. Through the sesamin prevent group(50mg/kg bw) and treatment group(50、100mg/kg bw) interventions, the triglyceride and cholesterol was decreased sig-nificantly, the serum HDL-C was increased significantly, and the lipids levels wasrepaired and restored.3. The study on mechanism was showed that:(1) Sesamin could decrease themalondialdehyde content, increase the total anti-oxidative capacity in, improve theoxidation resistance in liver and kidney organization.(2) Sesamin could lowerglutarnic oxaloacetid transaminase and glutamate pyruate transaminase content ofserum, reduce the extent of the liver fat disease and protect the liver cell.(3) Se-samin could improve the proportion of the serum free unsaturated fatty acids asoleic acid (C18:1) and linoleic acid (C18:2), reduce the proportion of saturatedfatty acid as palmitic acid (C16:0). It had a positive effect on regulating the serum free fatty acids metabolic disorders.(4) Sesamin could also descend gene expres-sion of diacylglycerol acyltransferas1and increase gene expression of camitinepalmitoyltransferase I which is the key enzyme of lipid metabolizing. It improvedthe mouse liver function of lipid metabolism.In conclusion, sesamin have preventive and curative effects on nonalcoholicfatty liver disease, the mechanism is relevant to control the lipid levels, free fattyacid composition, antioxidation, inhibit hepatocyte damage and the gene expressionof key enzyme in lipid metabolizing.
