Objective:Preliminary study the relationship between VEGF and HER2gene expression level in peripheral blood and Traditional Chinese Medicine syndromes in gastric carcinoma. To analyze the changes of VEGF, HER2gene expression level before and after the operation in gastric carcinoma. To analyze the association between VEGF, HER2gene expression level in peripheral blood in gastric carcinoma and clinicopathological characteristics.Methods:This study enrolled120cases of gastric carcinoma patients, and detect VEGF, HER2gene expression level by real time-polymorphisms chain reaction(RT-PCR) after respectively collecting2mL peripheral blood on preoperative and postoperative, to analyze the association between syndrome of gastric carcinoma and VEGF, HER2gene expression level in peripheral blood, and to analyze the association between VEGF, HER2gene expression level and clinicopathological characteristics. There are30cases of healthy subjects in the control group. Then statistically analyze the data by SPSS17.0.Results:1.The frequencies of TCM syndrome of gastric carcinoma:deficient cold of spleen and stomach>incoordination between liver and stomach>blood type drug resistance>phlegm-damp coagulation>deficiency of both qi and blood>yin-insufficiency due to stomach-heat.2.The VEGF, HER2gene expression level before the operation is higher than that of healthy control group, there is significant statistical difference between them (P<0.05).3.Gastric carcinoma in peripheral blood VEGF gene postoperative expression levels were obviously decreased, preoperative and postoperative VEGF gene expression levels had significant difference(P<0.05). Gastric carcinoma in peripheral blood HER2gene postoperative expression levels were obviously decreased, preoperative and postoperative HER2gene expression levels had significant difference (P<0.05).4.Gastric carcinoma preoperative of VEGF gene expression level, blood type drug resistance>yin-insufficiency due to stomach-heat>deficiency of both qi and blood>deficient cold of spleen and stomach>phlegm-damp coagulation>incoordination between liver and stomach, there are significant statistical differences between blood type drug resistance and deficient cold of spleen and stomach(P<0.05), between blood type drug resistance and phlegm-damp coagulate-on(P<0.05), between blood type drug resistance and liver and incoordination between liver and stomach(P<0.05); there is not significant statistical difference in any two groups (P>0.05).5.Gastric carcinoma preoperative of HER2gene expression level, phlegm-damp coagulation>yin-insufficiency due to stomach-heat>deficient cold of spleen and stomach>blood type drug resistance>deficiency of both qi and blood>incoordination between liver and stomach, there are significant statistical differences between phlegm-damp coagulation and deficient cold of spleen and stomach(P<0.05), between phlegm-damp coagulation and blood type drug resistance(P<0.05), between phlegm-damp coagulation and deficiency of both qi and blood(P<0.05), between phlegm-damp coagulation and incoordination between liver and stomach(P<0.05); there is not significant statistical difference in any two groups (P>0.05).6.There are statistical differences between syndromes in gastric carcinoma and TNM staging(P<0.05); deficient cold of spleen and stomach and incoordination between liver and stomach are more common in stage Ⅰ, stage Ⅱ gastric carcinoma, blood type drug resistance, phlegm-damp coagulation and deficiency of both qi and blood are more common in stage Ⅲ, stage IV gastric carcinoma.7.The VEGF gene expression level before the operation of gastric carcinoma is not related to age, gender, tumor location, differentiate degree, extension, lymphaden metastases, TNM staging(P>0.05). The HER2gene expression level before the operation of gastric carcinoma is not related to age, tumor location, extension, lymphaden metastases, TNM staging(P>0.05).8.The HER2gene expression level is related to gender and differentiate degree. There is significant statistical difference in the HER2gene expression level before the operation of gastric carcinoma(P<0.05), cardia part is higher than the gastric body and antrum. Not invaded the subserosa to the invasion of the subserosa, the deeper of the infiltration, the higher the expression level of HER2gene, there is significant statistical difference in them (P<0.05).Conclusion:1.There are different in VEGF, HER2gene expression level of TCM syndrome, therefore those points out that VEGF, HER2gene can be used as quantitative indicator to discern TCM syndrome differentiations of gastric carcinoma.2.The VEGF, HER2gene expression level before the operation is higher than that of healthy control group, which points out that VEGF, HER2gene can be early screening indexes of gastric carcinoma.3.Early gastric carcinoma with deficient cold of spleen and stomach and incoordination between liver and stomach, the remaining card type is more common in advanced gastric carcinoma.4.Gastric carcinoma in peripheral blood VEGF, HER2gene expression levels are obviously difference in preoperative and postoperative.Therefore, those points out that the VEGF, HER2gene can be used as effective indexes to estimate the condition assessment of gastric carcinoma, to provide some guidance of molecular targeted therapy and prognosis; that can be used as an effective indicator for screening high-risk patients.
