Objective:To describe the expression levels of RhoC protein in primary hepatocellular carcinoma(HCC);and investigate the role of RhoC protein in genesis and development,metastasis and recurrence,prognosis of HCC;and to explore the possibility of targeted therapy.Method:To compare the expression levels of RhoC between90cases of HCC tissues and corresponding adjacent tissues,combined with the pathological characteristics,the relationship between the expression of RhoC and VEGF were analyzed.There distinctions of expression in different pathologic tissues were studied.The multivariate COX proportional hazards model was used to analyze the prognositic recurrence risk factors.Results:1. RhoC protein staining positive signal was pale-brown or tan, mainly localized in the cytoplasm. Its expression in HCC tissues was significantly higher than the adjacent noncancerous normal tissue (p<0.01)2. According to the clinical data, with the gradual increase of RhoC expression, VEGF expression was increased gradually. In hepatocellular carcinoma, the expression of VEGF and RhoC expression was a significant positive correlation (rs=0.725,P<0.01)3. The expression of RhoC protein was not correlated with the gender, age, tumor size, AFP expression levels, HBV infection,but it was correlated with nodules number histological differentiation, portal vein invasion, lymph node metastasis and the envelope.4. Cox proportional hazards model, multivariate analysis showed that lymph node metastasis, portal vein invasion, no capsule, overexpression of RhoC indicates a poor prognosis, is an important independent risk factor for the impact of tumor recurrence and metastasis. Conclusion:The high expression of RhoC in hepatocellular carcinoma, involved in the genesis and development of liver cancer, liver cancer invasion and metastasis; the RhoC expression could up-regulate VEGF expression,and make tumor prone to blood transfer, play an important role in the invasion and metastasis of hepatocellular carcinoma; The RhoC with HCC free survival after surgery have a significant correlation,RhoC can be used as a prognostic indicator, and even can be used as a cancer treatment, newtargets.
