Primary Hepatocellular Carcinoma(PHCC) is a kind of malignant tumor whose prognosis is extremely bad. According to the statistics of USA, the 5 year survival rate of PHCC is 6 percent. At present, surgical operation is the most major treatment to it. Although the prognosis of PHCC has already improved along with the advances of the diagnostic radiology and the surgical techniques, the relapse rate after operation is still very high. For this reason, the relapse after operation has became the key problem which influences patient's prognosis. It is still not realistic depending merely on the improvement of surgical mode to bring about a striking treatmental effect. So, we must make a thorough study of PHCC's origin and development mechanism and look for some new treatmental methods.Rho protein which possesses the activity of GTP enzyme plays an important role in signal conduction route among cells, and also plays important roles in the movement of cells, the proliferation of cells, the death of cells, the transcription of cells, the transformation of cells and the infiltration and metastasis of malignant tumor cells. Rhoc is a member of rho gene family. In use of the technique of DNA chips, Clark and his colleagues examined the gene expression of the polygeneration tumor cell stub which does not metastases often and its derivative-tumor cell stub which metastases often. They discovered that the latter has a high gene expression. For the first time, their study confirmed that RhoC plays an important role in the metastasesing process of tumor cells. After analyzing the RhoC expression characteristics of 164 benign and malignant breast tumors, Kleer thought RhoC is the metastasesing signal of breast cancer. Some recent studies found that RhoC participates in the signal conduction among cells, the proliferation of cells and the regulation of actin's movement. Now, studies about the relation between RhoC and the relapse and metastasis of PHCC are very few.In this study, immunohistochemical method was used to detect the expression of RhoC protein in PHCC. Combinated with the analysis of clinical pathology characteristic and follow-up data, we investigated RhoC's function in PHCC's development and its possible mechanism tooffer a kind of measure for assessing the ability of tumor cell's invasion and metastasis, and also offer some theoretical basis for further studying PHCC's occurance and development mechanism and finding some new treatmental means.Material and Methods43 cases with PHCC operated in SRRSH were all confirmed by pathology from 2000 to 2002, including 25 males and 18 females. The average age of them was 53 years old. Grading them accoding to Edmondson pathologic standard: 13 cases belong to grade 1, 10 cases belong to grade 2, 14 cases belong to grade 3 and 6 cases are belong to grade 4.Follow-up: by telephone investigation, letter investigation and combinated with out-patient follow-up, we had investigated 36 patients with PHCC since their operation from the date of their operation to Oct, 2004 about whether they were alive, their survival time and their death reasons.Adopting the group method of S-P immunity, dilute anti-RhoC polyclone antibody with PBS to 1 percent and replace the first antibody with PBS to make it as negative contrast. To defined it as positive standard emerging light brown grain or brown grain in cytoplasm.According to the grade standard of Shimizu immunity, group as following: under high power lenses, shallow de strength marks 1, moderate dye strength marks 2 and deep dye strength marks3; no dye positive area marks 0, dye area less 1/3 marks l,dye area from 1/3 to 2/3 marks 2 and dye area more than 2/3 marks 3. add the above two as synthesized dye strength grade: mark 0 as "_", mark 1-2 as weak positive "+",mark 3-4 as positive"++" and mark 5-6 as strong positive"+++".
