The Effect Of On Cardic ACE2and ACE Protain Expression In Rat Myocardial Ischemia Treated By Losartan

Objective:To investigate the effect of on cardic ACE2and ACE protain expression in Rat Myocardial ischemia treated by losartan. Methods:MI modle was established by ligation of the anterior descending coronary artery in male Spague-Dawly (SD) rats. Twenty-four hours after the procedure, the36surviving rats were grouped randomly as mode group (n=6,2grops), losartan large dose treatment group and losartan low doses treatment group (n=6,4grops), Sham operated rats were selected randomly as non infarcted controls (n=12). Twenty-four hours after the procedure treat rats with losartan for losartan grops and physiological saline for mode grop with sham grop.14days and28days after treatment with the drugs by gastric gavage, the relative expression of myocardium ACE and ACE2protain was assessed by western-blot. Results:The protain expression of ACE and ACE2in mode group were significantly increased compared with sham group (P<0.01). The protain expression of ACE in28days losartan groups was markedly attenuated compared with mode group (P<0.05) except14days losartan groups. In contrast, losartan had significient effect on cardiac ACE2protain leval, which remained increased at the same levels in all areas of the MI rat heart (P<0.05). The ACE2protain leval of myocardic the losartan treated myocardial ischemia (MI) rats significantly increased compared with the non-treated ones (P<0.05).28days large dose losartan treatment myocardial ischemia (MI) rats significantly increased compared with the non-treated ones (P<0.05). The ACE protain leval of myocardic the28days large dose losartan treatment myocardial ischemia (MI) rats significantly decreased compared with the non-treated onesand with14days losartan low dose treatment group (P<0.05). The ACE protain leval of myocardic the losartan treated myocardial ischemia (MI) rats significantly decreased compared with the non-treated ones (P<0.05).Treatment of losartan can decrease the ratio of ACE to ACE2. Conclusion:The effect of enough losartan on the myocardial ischemia and regression of myocardic ischemia is related to the increase of ACE2protain expression creaseand decrease of ACE protein expression.
