Objectives1. To explore the change of ACE2—Ang-(1-7)—MAS path in the heart of SHR. 2. To explore effect of Losartan on ACE2—Ang-(1-7)—MAS path in SHR Methods24 30-week-old spontaneous hypertension rats and 12 Wistar rats of the same age were divided into 3 groups randomly: 1.WKY group, which were fed distilled water one time per day; 2.control group, which were fed distilled water one time per day; 3.SHR-Losartan group, which were fed Losartan 30mg/(kg.d); After 12 weeks of treatment, all rats were drawn for detection, including: 1.The level of PRA, AngII(including myocardial Angll level), Ang-(1-7)(inclouding myocardial Ang-(1-7) level) in blood serum; 2.Left ventricular end diastolic/systolic diameters (LVEDD/LVESD), thinknesses of interventricular septum (IVS) and post wall (LVPW) were measured at paprasternal left ventricular long-axis view with M-mode echocardiography, and mitral valve blood folw spectrums were recorded at apical four-chamber view with pulse-Doppler echocardiography; 3.Myocardial pathologic and ultrastructural changes were observed. 4.The mRNA expression levels of ACE, ACE2 and MAS were detected by RT-PCR respectively. 5.The protein levels of left ventricular ACE2,ERK1 ,ERK2 were detected by the immunohistochemical methods.
