| Objective:To analysis chemical composition of essential oils from different parts,orthogonal test design for optimization of the extraction of total flavoniods fromAgastache rugosa, and investgate its antibacterial activity. Meanwhile, explore actibactialchemical compisitons and mechanism of antibacterial activity, which will providrereferences for quality standard, clinical medication, rational and effective use of resources.Methods:①The different parts of A. rugosa was identified by morphological,histological identification, chemical qualitative and microscopic identification methods;②The essential oils of A. rugosa from Xinjiang, in China, were extracted by the methodof hydrodistillation and the chemical composition of the essential oils from A. rugosa, thedried flower and leaf, have been investigated by gas chromatography (GC) and gaschromatography-mass spectrometry (GC–MS);③Obtained best extraction conditions oftotal flavonoids of A. rugosa from Xinjiang through single factor exploration andorthogonal test design (L9(3)4);④Determined antibacterial activities of essential oilsfrom different parts, total flavonoids, water extracts and ethanol extracts by serial dilutionmethod and agar diffusion method (modified Kirby-Bauer method);⑤Utilized96-holemethod and calcutated inhibition rate to analysis various samples effect on bacteria andbiofilm formation. Results:①The yields of essential oils from the dried flower, stem andleaf based on the dry weight of the plant were0.29%,0.02%and0.57%(w/w),respectively.②The flower oil was revealed the presence of21components, representing99.78%of the total oil. The major compounds of the flower oil were oxygenated terpenes(35.4%), estragole (29.5%), p-Menthan-3-one (19.2%), and monoterpenes (8.8%).Twenty-six components accounting for99.62%of constituents of leaf oil were identified,and the main compounds of the oil were p-Menthan-3-one (48.8%), estragole (20.8%),monoterpenes (8.3%), oxygenated terpenes (5.6%).③Derermination of best extractionconditions of total flavonoids of A. rugosa from Xinjiang: solvent concertration, ratio ofsolvent to raw material, extraction time, extraction times were80%,20:1,30min,2,respectively. The content of total flavonoids is33.46mg/g.④The essential oils fromdifferent parts, total flavonoids, water extracts and ethanol extracts of A. rugosa fromXinjiang have certain antibacterial activities for Staphylococcus aureus,Escherichia coli, Blastomyces albicans, Streptococcus mutants. Conclusions:The development of thisstudy is helpful to find new medical values and elucidate mechanism, which will offerexperimental basis for select new antibacterial medicines from traditional Chinesemedicine and ethnic medicine, and make sufficient fundamental work for development ofindependent intellectual property of medical research. It play important role in deeperexploration of medicinal material and development of traditional Chinese medicine andethnic medicine industry. |
