objective: to explore Thl/Th2change trends in COPD bystudy on expression changes of STAT4, STAT6in acute exacerbationstage and stable stage patients of patients with COPD, and while toinvestigate the relationship of lung function in patients with COPD andSTAT4, STAT6and its clinical significance. Methods: Staining theperipheral blood mononuclear cells in72patients with COPD withSTAT4and STAT6Rabbit anti-Human Polyclonal Antibodies byimmunocytochemical method, and20cases of normal people as control,and then detected and analysed on their expression. Results:1:Expression of STAT4in PBMC of COPD patients’ blood wassignificantly increased, there were not obvious difference in acuteexacerbation stage and stable stage, the expression of STAT4in FEV1≥50%predicted group is higher than FEV1<50%predicted group, thedifference was statistically significant, it suggest that there werecorrelation between STAT4abnormal activation and the severity ofpulmonary function decline in patients with COPD but no correlation oftheir staging;2: The expression of STAT6of PBMC in acuteexacerbations COPD patients was increased, compared with the controlgroup was statistically significant, but in FEV1≥50%predicted groupand FEV1<50%predicted group, the expression of STAT6had no stable COPD patients with the control group, no difference, it suggest thatthere were significant between increased expression of STAT6andCOPD stage but no correlation with the severity of lung function decline;3: The expression of STAT6was increased significantly in acuteexacerbations COPD patients than STAT4, the STAT4/STAT6ratiodecreased, the differentiation of Th cells present Th2advantage condition;the expression of STAT4increased significantly higher than that ofSTAT6in stability period, the STAT4/STAT6ratio was significantlyincreased, Th cell differe-ntiation has the advantages of Th1state.Conclusion: STAT4abnormal activation was related to the severity ofdecreased lung function of COPD patients, and increased expression ofSTAT6was related to COPD staging, there were Thl/Th2cell ratio imba-lance in acute exacerbation stage and stable stage, acute exacer-bationof COPD who exist mainly to changes in the direction of Th2, stableperiod were dominated by Thl direction changes.
