Objective Schistosoma japonicum infection the obese mice in the chronic phase can improve the insulin resistance. In this study, Insulin resistance in C57BL/6J mice was induced by high fat diet, further to investigate the possible mechanism above the obese mouse infected with Schistosoma japonicum with the liver insulin resistnce.Methods 36 male C57BL/6J mice were randomly assigned into 3 groups:normal control (NC, n=12) group, high-fat diet (HF, n=12) group and high-fat diet with schistosoma japonicum infected (HSj, n=12) group. They were fed with standard diet or high-fat diet group respectively, and mice in HSj group were infected schistosoma japonicum. Specimen was collected separately at end of 6th and 12th week after high-fat diet. The levels of fasting blood glucose (FBG) and fasting plasma insulin (FINS) were detected first, in order to get the result of homeostasis model assessment index for insulin resistance (HOMA-IR). interferon-γ(IFN-γ),interleukin-4(IL-4) and singal transductor and activator of transcription-4(STAT-4) singal transductor and activator of transcription-6 (STAT-6) were detected by ELISA and Immunohistochemical method.Result1. Sixth weeks after infection of Schistosoma japonicum cercariae, the body weight of the mice in HF group and HSj group gained 20% compared with the average body weight in NC group, so the obesity model induced by HF diet was established successfully. By the end of 6 weeks, the weight of mice in NC group were lower than that in HFgroup (P<0.01) and HSJ group (P<0.05); By the end of 12 weeks, the weight of mice in HF group were higher than that in NC group (P<0.01), the weight of mice in HSJ group were lower than that in HF group (P<0.01)2. By the end of 6 weeks,the levels of HOMA-IR and FINS in mice from HF group were higher than NC group(P<0.01)。the levels of HOMA-IR and FINS in mice from HSJ group were higher than NC group(P<0.01); By the end of 12 weeks, the levels of HOMA-IR and FINS in mice from HF group were higher than NC group(P<0.01).the levels of FINS、FBG and HOMA-IR in mice from HSJ group were lower than HF group(P<0.05); By the end of 12 weeks,the levels of FBG in the mice from the group HSJ were lower than the end of 6 weeks(P<0.05),the levels of FINS in the mice from the group HF were higher than the end of 6 weeks(P<0.01).3. By the end of 6 weeks, the levels of IL-4 and IFN-y in the mice group of HSJ were higher than NC group(P<0.01).The levels of IFN-y in the mice group of HF were higher than NC group(P<0.01).The levels of IL-4 in the mice group of HSJ were higher than HFgroup(P<0.01); By the end of 12 weeks,the levels of IFN-y and IL-4 in the mice from the group HF were higher than NC group(P<0.05).The levels of IFN-y and IL-4 in the mice from the group HSJ were higher than NC group(P<0.01).The levels of IL-4 in the mice from the group HSJ were higher than the HF group(P<0.01);By the end of 12 weeks,the levels of IFN-y and IL-4 in the mice from the group HF were higher than the end of 6 weeks (P<0.05)4. By the end of 6 weeks, the levels of STST4 and STST6 in the mice group of HF were higher than NC group(P<0.01). The levels of STAT4 and STST6 in the mice group of HSJ were higher than NC group(P<0.01); By the end of 12 weeks, the levels of STAT4 and STAT6 in the mice group of HF were higher than NC group(P<0.05). The levels of STST6 in the mice group of HSJ were higher than HF group(P<0.05); By the end of 12 weeks, the levels of STAT6 in the mice group of HSJ were higher than the end of 6 weeks (P<0.05)Conclusion1. The mouse with high fat diet of obesity with characteristics of insulin resistance.2. The inflammatory cytokines IFN-γ and STAT4 were increased in the obese mice liver tissue,the ant-inflammatory cytokines IL-4and STAT6 were reduce, so the obese mice with insulin resistance may exist Thl-type cell polarization.3. Schistosoma japonicum infection in advanced might promote Th2 cells polarization in liver tissue of obese mice, reduce inflammation and ameliorate insulin resistance.
