Objective:To investigate the expression and significance of hypoxia-inducible factor-la and bcl-2in rats after MCAO at different time periods, assessed the relationship between HIF-1α and Bcl-2, then provide the experimental basis to cerebral infarction therapeutic targets. Methods:The healthy male and female SD rats which weight250-320g, were randomly assigned to three groups: groupA(control group,N=4), groupB (surgery group,N=16) and groupC(sham operation group, N=16)。 Then, we divided the group B and C to8small groups at different time periods(0.5h,2h,3h,6h,12h,24h,78h and7d) MCAO model was induced by modified Longa; s method. Then rats were subjected to a neurobehavioral deficit test, only the rats who got1-3scores can be chosen。 The ELISA method was used to measure the OD values of HIF-1α and bcl-2. Then draw the standard curve by standard concentration.Finally, we calculated HIF-la and bcl-2expression levels by standard curve and used analysis of variance to evaluate all data at different time periods and different treatments. Surgery rats were randomly selected to take of TTC staining after the head perfusion by saline to judge if the model is succesful. Results:1) Neurobehavioral deficit scores:surgical group scored1.63±0.554,Sham operation group and control group scored0point, the difference was statistically significant (P<0.05);2) HIF-1α expression: surgical group increased significantly higher than Sham operation group and sham group if we not considerate the difference cased by time changing。 Surgical group increased significantly higher than Sham operation group and Sham group at0.5h、2h、3h、6h、12h (P<0.05)。The change between different time points in surgical group:HIF-1α expression levels at12h was statistically significant than the other time points except at6h, HIF-1α expression levels among0.5h、2h、24h、3d、7d were unstatistically significant(P<0.05).3)Bcl-2expression results:surgical group increased significantly higher than Sham operation group and sham group if we not considerate the difference cased by time changing; Surgical group increased significantly higher than Sham operation group at6h、12h、24h (P<0.05), surgical group increased significantly higher than Sham group at2h、3h、6h、12h、24h、3d (P<0.05). The change between different time points in surgical group:Bcl-2expression levels at6h、12h、24h was statistically significant than the other time points, the expression levels at6h、12h were statistically significant than24h, and the other time perids were not statistically significant to each other (P<0.05)。4) TTC staining:the area right middle cerebral artery supplied was stained white, which imply the infarct areas. Conclusion:1) Modified MCAO model is an good animal model for focal cerebral infarction;2) The cerebral neurobehavioral deficit scores of surgery group was higher than sham operation group, take of TTC staining judge if the model was succesful。3) HIF-1α expression started from0.5hour,12hours reached a peak,then decreased。 HIF-1α play an important role after rat acute cerebral infarction.4) Bcl-2expression started from6hour,12hours reached a peak then decreased,3day reached normal level。 As a apoptosis gene; it involved in the neuroprotection of cerebral infarction.5)The HIF-1α expressed earlier than Bcl-2, but reached peak at the same time and then decreased, theycould have close relationship。...
