Objective To detect the expression of twist and E-cadherin in normal cervical tissues, cervical intraepithelial neoplasia(CIN), and cervical cancer, and to investigate the role of twist and E-cadherin in the occurrence, invasion and metastasis of cervical cancer.Methods Immunohistochemical method was employed to detect the expression of twist and E-cadherin in20cases of normal cervical tissues,30cases of CIN, and70cases of cervical cancer. To explore the relationships between the two proteins expression and age, clinical stage, pathological type, histological differentiation, the depth of tumor invasion and lymph node metastasis of cervical cancer.Results1. Twist was mainly localized in the nucleus. E-cadherin in CIN and cervical cancer was mainly localized in the cell membrance and in normal cervical tissues was only localized in the cell membrance.2. The positive expression rates of twist were74.29%、36.67%and15%in cervical cancer, CIN and normal cervical tissues, respectively. The positive expression rate of twist in cervical cancer was significantly higher than that in normal cervical tissues and CIN. The positive expression rate of twist in cervical cancer was associated with the depth of tumor invasion and lymph node metastasis, but not with age, clinical stage, pathological type, histological differentiation.3. The positive expression rates of E-cadherin were38.57%、66.67%and80%in cervical cancer, CIN and normal cervical tissues, respectively. The positive expression rate of E-cadherin in cervical cancer was significantly lower than that in normal cervical tissues and CIN. The positive expression rate of E-cadherin in cervical cancer was associated with the depth of tumor invasion and lymph node metastasis, but not with age, clinical stage, pathological type, histological differentiation.4. The expression of twist was negatively correlated with that of E-cadherin in cervical cancer.Conclusion 1.The up-regulation of twist and down-regulation of E-cadherin may promote the pathogenesis、invasion and metastasis of cervical cancer.2.Twist may play as the promoter of tumor’s invasion and metastasis associated with down-regulation of E-cadherin.
