Background:miRNA-29a which was cloned from Human Hela cells in2001is the non-coding transcribed single copy gene cluster and is located in chromosome7q32.2. Recently, the study of the relationship of miRNA-29a and abdominal aortic aneurysm(AAA) is lack, and all of the study are the relationship of miRNA-29a expression and AAA tissues. But, the study of the expression and significance of miRNA-29a in the serum of AAA has not been reported.Objective:To study the expression of miRNA-29a in the serum of patients with infrarenal abdominal aortic aneurysms(IAAAs), and to investigate the relationship between the expression and risk factors such as Aneurysm size, age, sex, smoking, hypertension and intraluminal thrombus.Methods:Twenty cases with an average age of63.1±14.2years of IAAA patients’serum specimen were collected from November2011to April2012in Xiangya hospital. At the same time, twenty cases of normal volunteers’with an average age of64.8±12.5years serum specimen were collected from Health Evaluation Center in Xiangya hospital as controls. RT-PCR testing was respectively performed on the serum samples in two groups for detecting miRNA-29a relatively expression, then comparing the difference between the two groups. The AAA group was respectively divided into two groups according to the factors such as aneurysm size, age, gender, medical history and intraluminal thrombus, then comparing the difference between the two groups.Results:Regression analysis showed no significant influence of age or sex between AAA group and the control group. RT-PCR could detect the expression of miRNA-29a in human serum. Decreased expression levels of miRNA-29a were observed in AAA group comparing with controls(2.40±0.72vs6.96±1.56)(P<0.001). The AAA group were divided in two groups according to aneurysm size:AAAD1:aneurysm size <5.5cm and AAAd2:aneurysm size≥5.5cm. The expression of miRNA-29a between the two group exist significant differences(2.90±0.58vs1.91±0.44)(P<0.001), and with the increase of aneurysm size, The expression of miRNA-29a gradually decreased. The AAA group were divided in two groups according to age:AAAA1:age<65years and AAAA2:age≥65years. The expression of miRNA-29a between the two group do not exist significant differences (2.58±0.73vs2.29±0.72)(P>0.05). The AAA group was divided in two groups according to sex: AAAG1:Female and AAAG2:Male. The expression of miRNA-29a between the two group do not exist significant differences yet (2.30±1.01vs2.43±0.66)(P>0.05). The AAA group were divided in two groups according to smoking or not:AAAS1:smoking and AAAS2: non-smoking. The expression of miRNA-29a between the two group exist significant differences, decreased expression levels of miRNA-29a were observed in AAAS1group comparing with AAAS2(2.08±0.45vs2.81±0.80)(P<0.05). The AAA group were divided in two groups according to hypertension or not:AAAH1:hypertension and AAAH2:non-hypertension. The expression of miRNA-29a between the two group do not exist significant differences (2.52±0.78vs2.23±0.38)(P>0.05). The AAA group were divided in two groups according to having obvious intraluminal thrombus or not:AAAT1:intraluminal thrombus and AAAT2: non-intraluminal thrombus. The expression of miRNA-29a between the two group exist significant differences, decreased expression levels of miRNA-29a were observed in AAAT1group comparing with AAAT2(2.16 ±0.57vs2.71±0.79)(P<0.05).Conclusion:1. The AAA patient’s serum has decreased expression levels of miRNA-29a comparing with average person.2. In human’s serum, with diameter of the abdominal aortic increasing, the expression levels of miRNA-29a decreases.3. The expression levels of miRNA-29a in AAA patients’serum exist differences which involving in aneurysm size, smoking or not and having obvious intraluminal thrombus or not but having nothing to do with age, sex and hypertension history.
