Objective:To establish new golden hamster model for atherosclero-sis and explore the effects of dyslipidemia, diabetes and low shear stress on atherosclerosis.Materials and Methods:100male Syrian golden hamsters, weighing between95-100g, were randomized into5groups with20in each group:non-treated group (N), hyperglycemic group (G),hyperlipid-emic group (L), combined hyperlipidemic/hyperglycemic group (L+G) and combined hyperlipidemic and low shear stress group (L+L), among which, golden hamsters in G group and L+G group were injected with streptozotocin (STZ). After4weeks of high fat diet feeding, hamsters in L+L group were ligated on left internal carotid artery and external carotid artery, only with occipital artery untreated. And the following aspects were explored:l、to determine if one single does of streptozotocin (STZ) would produce hyperglycemia similar to diabetes2、 whether the following factors were associated with the development and acceleration of atherosclerosis.①variations of plasma glucose, triglyceride, total cholesterol and HDL-C.②TC/HDL-C and TG/HDL-C ratio value.3、To get a better understanding of the effects of low shear stress on atherosclerosis by combining high fat diet.Results:1、Plasma glucose of the hamsters increased significantly after one IP injection of STZ and symptoms similar to human diabetes were observed.2、Lipidtriad of golden hamsters with atherosclerosis showed characteristics of increase of triglyceride level and small dense LDL level and decrease of HDL-C level at20week timepoint.3、The golden hamsters treated with STZ did not show insulin dependence and β cells continued insulin secretion which is similar to type2diabetes. Golden hamsters in L+G showed insulin resistance and insulin dysfunction at the same time.4、Obvious acceleration of atherosclerosis was observed in L+G and L+L, while golden hamsters only with hyperglycemia did not show acceleration of symptoms. Atherosclerosis is mainly associated with increase of TC and HDL-C level. When combined with increase of small, dense LDL particles and hyperlipidemia, atherosclerosis was more likely to be accelerated for oxidation caused by these substances.5、 Low shear stress combined with hyperlipidemia could accelerate the development of atherosclerosis.6、 The diameter of left carotid artery decreased by40%compared to right carotid artery in L+L group. Vessel diameter in L+L is the minimum among all the groups.Conclusion:1、 Golden hamster model for atherosclerosis was successfully induced in this experiment.2、 Combined hyperglycemia and hyperlipidemia as well as hyperlipidemia and low shear stress could accelerate the development of atherosclerosis.3、 Vascular morphology was validated combining imaging and anatomy, the diameters of blood vessels of golden hamster in atherosclerosis were smaller than those in other groups and the diameters of the vessels on the ligated side decreased by approximately40%comapared to those on the opposite side.
