Study of the relevance of signal transduction and activator of transcription STAT6gene+2964G/A polymorphism and Allergic rhinitis of Shandong Han population.Material and Methods:1Study object:According to the Diagnosis and treatment principles of allergic rhinitis (LanZhou2004), choose natural population of Shandong, Grouped according to uniform standards,104patients with AR and115healthy controls2STAT6G2964polymorphism detection.(1)Venous blood4ml (EDTA-Anticoagulant),Separation of serum. DNA was extracted By genomic DNA Extraction kit, PCR amplification and PCR amplification products were sequenced by Bio-Engineering Company. Then analysised the Gene polymorphism of the Sequencing results.(2) plasma IL-13level were determined by ELISA.(3) take the separated plasma Use the Mast detection system(Produced by Germany), TIgE and SIgE Positive allergens corresponding SIgE level which measured by skin prick test.(4) Statistical analysis:Process the data With the SPSS13.0statistical software.Results:1Significant differences in genotype were observed between AR group and normal control group. In AR group the frequency of GG in G2964A sites was higher than Control group (χ2=7.17, P=0.028;), the frequency of Allele G was higher than Control group (χ2=5.82, P=0.022)2For the same genotype, IL-13concentration in AR group was higher than that in control group. In the same group,IL-13concentration of G allele carriers was higher than that of control group(P<0.05).3There was no significant differences of TIgE concentration between AR group genotypes (P>0.05).4There was no statistically significant in SIgE of Dust mites and ragweed pollen between different genotypes of STAT6G2964A sites (P>0.05). Conclusion:STAT6gene2964G/A site is associated with Allergic rhinitis, this site may be an important candidate gene for AR, and can affect the plasma IL-13level...
