0bjective To investigate the baicalin’s protective effect on oxidation damage in A549providethe experimental basis for the development of baicalin health care at respiratory symptom andthe application of baicalin additive in chinese cigarette, human alveolar epithelial cells ofoxidative damage model was established, and protective effect on oxidation damage wasobserved.Methods Establish human alveolar epithelial cellsof oxidative damage model:A549cells weretreated with different concentrations of H2O2,and assayed at the indicated time points by MTT.After that, the best condition was determined,that the manufacture of oxidative damage model inA549. There were three groups:blank control, H2O2,H2O2+baicalin(0.05mg/L,0.1mg/L,0.2mg/L,0.4mg/L).Cell viability was evaluated by MTT assay and thelevel of MDA and SOD、NOS、GSH-PX in cell culture supernatant was measured withantioxidation testing cassete.Results1.Through this experiment,the best condition was determined,that the manufacture ofoxidative damage model in A549.The best condition was that the concentration of H2O2was200μmol/L and treating time was24hours.2.MTT assay results showed that H2O2decrease theviability of A549cells (P<0.05), the protein level decreased and oxidative damage index rose ongroup of H2O2. Baicalin inhibited the decrease of A549cell viability and protein induced byH2O2(P<0.05). The viability of SOD,NOS,GSH-PX and protein content increased significantly.Conclusion1. The model were made successfully in A549cells.2.Bacalin could inhibite theinjury caused by H2O2and would reduce the oxidative damage effect induced by H2O2(P<0.05).
