Objectives:To study the different concentrations of BaP to the impact of oxidation and antioxidation status of alveolar typeⅡcells and variation of antioxidant enzymes,to investigate BaP in cigarette smoke on oxidative damage mechanisms of the airway epithelial cells in order to offer cytotoxicity basis.Methods:A549 cells to simulate the human alveolar typeⅡcells as research subjects,the use of 0.1umol/1,1.0umol/1,10.0umol/1 BaP for 72 hours to continue treating A549 cells,MDA levels detected from the protein level,TAC,total SOD,MnSOD,Cu/ZnSOD and CAT activityand then use RT-PCP to detect the expression of MnSOD.Results:With the increased concentration of B(a)P, the levels of lipid peroxidation (MDA) in A549 cells increased in a concentration-depend manner. The total activity of SOD and the activity of MnSOD increased after 0.1μM B(a)P treatment, which have significant difference (P<0.05) compared to the control group. But the total activity of SOD and the activity of MnSOD decreased after 1μM and 10μM B(a)P treatment. The expression levels of MnSOD mRNA also have the same trend with the MnSOD activity. With the increased concentration of B(a)P, the catalase activity(CAT) also increased, which compared with the contrl group increased in 1.4,1.5 and 2.3 folds.Conclusions:B(a)P pollution lead A549 to increase levels of lipid peroxidation,and MDA was proportional to the B(a)P concentration,indicating that the higher the concentration,the more severe cellular oxidative damage.B(a)P pollution caused adaptive changes of antioxidant enzymes,TAC and CAT were up-regulated and total SOD and MnSOD activty were first increased and then dedcreased with increased B(a)P concentration.
