Objective:To explore the expression of miR-21and miR-143in cervical squamous cellcarcinoma and study the relationship of miR-21and miR-143with FIGO stage,histologic features, tumor size, differentiation, lymph node metastasis, neoadjuvantchemotherapy.Methods:1. Collected the clinical data of50cases with cervical squamous cell carcinoma andfound out the corresponding biopsy and paraffin-embedded tissue.2. Isolated cervical squamous cell carcinoma and adjacent tissues bymicrodissection technique with sterile syringe needles, extracted total RNA,detected the expression of miR-21and miR-143in cervical squamous cellcarcinoma by real-time quantitative PCR method.3. Correlation of relative expression level of those two microRNAs and FIGO stage,histologic features, tumor size, differentiation, lymph node metastasis,neoadjuvant chemotherapy were analyzed by statistical analysis.Results:1. The mean relative expression level of miR-21was significantly higher thannormal tissue and the difference was statistically significant(t=3.026, P<0.05);the mean relative expression level of miR-143was significantly lower thannormal tissue and the difference was statistically significant(t=2.515, P<0.05).2. The mean relative expression level of miR-21and miR-143in poor differentiatedgroups were1.924±0.182and0.722±0.075; the mean relative expression level ofmiR-21and miR-143in well differentiated groups were1.643±0.157and0.951±0.082. Expression of these two genes were statistical significance between two groups(t=2.248and t=2.197, P<0.05).3. The mean relative expression level of miR-21and miR-143in lymph nodemetastasis positive groups were1.937±0.137and0.761±0.085, the mean relativeexpression level of miR-21and miR-143in lymph node metastasis negativegroups were1.653±0.108and0.907±0.073. Expression of these two genes werestatistical significance between two groups(t=2.248and t=2.197, P<0.05).4. The mean relative expression level of miR-21and miR-143were no statisticalsignificance in FIGO stageⅠ group and FIGO stageⅡgroup(t=1.313andt=1.292, P>0.05).5. The mean relative expression level of miR-21and miR-143were no statisticalsignificance in keratinized group and non keratinized group(t=1.224and t=1.458,P>0.05).6. The mean relative expression level of miR-21and miR-143were no statisticalsignificance in large tumor size group and small tumor size group(t=1.552andt=1.415, P>0.05).7. The mean relative expression level of miR-21and miR-143were no statisticalsignificance in neoadjuvant chemotherapy group and non-neoadjuvantchemotherapy group(t=1.359and t=1.377, P>0.05).Conclusions:1. The expression of miR-21in cervical squamous cell carcinoma was significantlyhigher than normal tissue. MiR-21expression were significantly associated withdifferentiation, lymph node metastasis and were not significantly associated withFIGO stage, tumor size, neoadjuvant chemotherapy, histologic features.2. The expression of miR-143in cervical squamous cell carcinoma was significantlylower than normal tissue. MiR-143expression were significantly associated withdifferentiation, lymph node metastasis and were not significantly associated withFIGO stage, tumor size, neoadjuvant chemotherapy, histologic features.
