PART ONE EFFECT OF LONICERA EXTRACT FROMWATER SOLUTION ON THE TREATMENT TOOVALBUMIN-SENSITIZED BALB/C MICEObjective To study the effect of lonicera extract from water solutionon ovalbumin-sensitized BALB/c mice.Methods According to giving the different concentrations of loniceraextract from water solution,40ovalbumin-sensitized BALB/c mice wererandomly divided into group H (1g/ml as high concentration), group M(0.5g/ml as middle concentration), group L (0.25g/ml as low concentration),and positive control (hypersensitivity), negative control (normal). Thefootpad swelling reactions were used to assess for the OVA-induced delayedhypersensitivity. And mast cells in jejunum and mesentery in the mice weredetected by toluidine blue staining. The villus morphology in jejunum wereobserved by HE staining. The serum OVA-specific IgE and IgG1weremeasured by ELISA.The total IgA and OVA-specific IgA levels of intestinalmucus were measured by ELISA. Results All the mice presented acute diarrhea after challenged withOVA, and the pathological section of intestine in the mice showed thatdegranulated mast cells and the inflammatory reactions were serious. But thediarrhea in the mice were significantly improved after fed the high andmiddle concentrations of lonicera extract from water solution. And thedegranulated mast cells in jejunum and mesentery were markedly decreased.The inflammatory reactions in jejunum were relieved. The OVA-specific IgElevels of serum and OVA-specific IgA levels of intestinal mucus weremarkedly decreased. And the footpad swelling reactions were inhibited.Conclusion The study demonstrated that Lonicerae Japonicae Flosshowed effective to the hypersensitived mice by OVA-induced. The highconcentration of Lonicera extract from water was more effective than others. PART TWO EXPLORING THE EXPRESSION OF MUC1AND MUC2IN THE OVA-SENSITIZED MICEObjective To explore the expressions of MUC1and MUC2in theOVA-sensitized mice.Methods16female BALB/c mice with56weeks were randomlydivided into hypersensitivity and normal groups. The animal model of foodallergy was established with OVA in hypersensitivity group, then the OVA specific IgE、IgG1levers of serum and the total IgA and OVA-specific IgAlevels of intestinal mucus were measured by ELISA. The mast cells inJejunum and mesentery in the mice were detected by toluidine blue staining;The villus morphology and mucins in stomach and jejunum wererespectively observed by HE staining and AB/PAS. Immuno-histochemicalSP-staining method was performed to detect the expressions of MUC1andMUC2in stomach and jejunum.Results The OVA specific IgE、IgG1levers of serum and the totalIgA、 OVA-specific IgA levels of intestinal mucus were increased inOVA-sensitized mice. And the degranulated mast cells in jejunum andmesentery were increased, the inflammation reactions in jejunum wereserious, the goblel cells and mucins were increased in jejunum. The MUC1in stomach and jejunum were expressed in OVA sensitized mice, but not innormal mice; then the MUC2in jejunum of two groups were expressed, butit was significantly higher in OVA sensitized mice than that in normalmice(P<0.05).Conclusion The up-regulation of MUC1and MUC2may be involvedin progression in food allergy, especially MUC1.
