Part oneTherapeutic Evaluation of the Lonicera Extract from Water Solution Used in the Ovalbumin-Induced BALB/c Mice【Objective】 Investigate the therapeutic effects of the Lonicera extract from water solution (Lonicera ext) in the ovalbulmin-induced BALB/c mice.【Methods】 Female BALB/c mice aged at 6 weeks fed on the ovalbulmin-free diet, were randomly divided into 4 groups by the intragastrical treatments after being sensitized and challenged with ovalbulmin (OVA), including group H treated with 100% of the Lonicera extract from water solution, group M with 50%, group L with 25%, and group Ch treated with normal saline as the positive control. Meanwhile, the mice in group NS without sensitization and challenge were set as the negative control.(1)The effects of the Lonicera ext on the OVA-specific IgE and IgG1 levels in serum in the OVA-mediated immediate allergic mice were evaluated by the heat-unstable and the heat-stable PCA (Passive Cutaneous Anaphylaxis) activity respectively; (2)The histological examinations with either HE or toludine blue stain were performed in the mice; (3)The levels of histamine in gut in the mice were assayed with a fluorescent method;(4)The footpad tumefaction reactions on the OVA-induced delayed allergic murine model were assessed to investigate the effects of the Lonicera ext.【Results】 (1)The OVA-induced heat-unstable reactions were inhibited after the mice with OVA allergy were treated by gavage with 100% or 50% of the Loncera ext for consecutive 10 days.(2)The significant inflammatory reactions, such as the accumulation of the lymphocytes and plasmacytes in lamina propria (LP) and the focal necrosis and shedding of the epithelial cells in intestinal villi, were inhibited in the mice of group H and M;(3)The accumulated and degranulated mast cells in LP were statistically reduced in the mice by gavage with 100% or 50% of the Loncera ext, concomitant with the increased percentage of the intact mast cells.(4)The release of histamine in gut in the mice of group H and M was significantly reduced.(5)The footpad tumefaction reactions induced in the allergic mice were significantly inhibited after given the solution of the Lonicera ext. 【Conclusions】 The therapeutic effects of the Lonicera ext on the OVA-induced allergic mice model were studied first, and the data showed that the Lonicera ext could be used for food allergic disorders.
