Objective:To investigate the association of serum angiotensin converting enzyme activity and angiotensin converting enzyme gene polymorphism with cerebral infarction, vascular dementia and Alzheimer's disease.Methods:Using the polymerase chain reaction(PCR)-AFLP assay, angiotensin converting enzyme(ACE) gene polymorphism were analyzed in 62 patients with vascular dementia (VD), 38 patients with Alzheimer's disease(AD), 58 patients with cerebral infarction (CI) and 46 healthy controls. The serum activity of angiotensin converting enzyme in 47 patients with VD, 15 patients with AD, 43 patients with CI and 30 healthy controls were measured by means of capillary electrophoresis ultraviolet detection.Results:Compared with that of controls(22.62+5.44U/L), the serum ACE activity was singnificantly lower in Cl patients( 19.26 + 5.11 U/L), particularly in those patients with hypertension(18.42+5.20 U/L) and male ones(19.71+4.99 U/L). We did not find any association between ACE gene polymorphism and serum activity in patients with vascular dementia. Although frequency of II genotype and I allele seemed higher in AD group than those in the controls, Difference were not of statistical significance( x2=1.573 , P>0.05; x2=1.115 , P>0.05, respectively.).Conclusions:The serum ACE activity decrease in the event of ischemic stroke, and is related to the degree of cerebral tissue damage. There was no significant corelation between the 1/D polymorphism of ACE gene and vascular dementia. Further study of the association between ACE polymorphism and AD is needed.
