Study On The Active Constituents Of Cassia Seed

Semen Cassia has a good lowering blood lipid and blood pressure, improving eyesightand protecting liver effects and so on. For the sake of studying on effective constitutions oflowering blood lipid in Semen Cassia, this paper took it as research project, studied on thechemical constitutions of Semen Cassia by extracting, isolating and purifying them and themolecular structure of the effective constitutions was identified in a few different spectrumtechnique and method. These lay the foundations of the exploitation of Semen Cassia. Theresearch results are as follows:1. The extraction of effective constitutions in Semen CassiaWhether Semen Cassia needed to be ground before extracted was decided byexperiments in this paper. The result was that extraction efficiency was increased obviouslyafter grinding. To investigate that the factors such as the soak time and the ethanolconcentration affected extraction efficiency, the best conditions were optimized that one hoursoak time and70%ethanol concentration at last through single factor method in this paper.Adopting the best conditions, Semen Cassia (3kg) was extracted, the extractives (652.3g)were attained and the extraction efficiency was18.6%.2. The isolation and purification of effective constitutionsThe extended extractives were isolated to obtain petroleum ether (PE), ethyl acetate (EA),n-butyl alcohol, methanol and70%ethanol five extractive fraction through heating refluxextraction crude separation. In this experiment the chemical constitutions of PE fraction andEA fraction were studied. They were isolated further by gel column chromatography.3. Identification of the structure of pure compoundsSeven compounds were isolated in this study. Through1D NMR,2D NMR and massspectrum comparing with relevance literatures, and compared with Physcion, Chrysophanoland Emodin three reference substances by TLC we confirmed five compound: Chrysophanol(CS-2),1-desmethyl-obtusin(CS-4), Physcion(CS-6, CS-10and CS-17), Obtusifolin(CS-5andCS-13) and Aurantio-obtusin (CS-15). The structures of CS-8and CS-12are being analyzed.
