| Objective:To investigate the correlation between the expression of Ang2,HIF-1α,VEGFand Helicobacter pylori L-form infection and their effect on the biological behaviors ofgastric cancer,such as its angiogenesis,invasion and metastasis.Methods: Collected60cases of paraffin blocks of surgical specimens about gastriccancer and clinical pathological data, obtained30cases from the correspondingadjacent tissues (the junction of normal and cancer, adjacent group), and30cases ofnormal gastric mucosa margin (control group).(1) Immunohistochemical staining withElivision method was used to detect the expression of Ang2,HIF-1α,VEGF protein inthe above tissues.(2) Hp-L was examined in the above tissues by Immunohistochemicalstaining with Elivision method and Gram stain.(3) The expression of Ang2mRNA in30cases of fresh gastric cancer tissues and the normal gastric mucosas margin (controlgroup) were detected by semi-quantitative reverse transcriptase-polymerase chainreaction (RT-PCR).Results:(1) The positive rate of the expression of Ang2,HIF-1α,VEGF in gastriccarcinoma was higher than their control group(P<0.05). Their expression had a definitecorrelation (r=0.358, r=0.839, r=0.400,P<0.05); and was correlated with histologicaldifferentiation,lymph node metastasis and TNM stage(P<0.05).(2) The result of Hp-Linfection was observed by Immunohistochemical stain,which was consistent with it byGram’s staining(P<0.05). The positive rate of Hp-L in gastric cancer, paracancerous andcontrol group was68.3%,56.7%,20.0%. The positive rate in gastric cancer comparedwith the control group had statistical significance(P<0.05), adjacent compared with thecontrol group also had statistical significance (P<0.05), but gastric cancer comparedwith adjacent was not statistically significant(P>0.05).(3) In gastric cancer, theexpression of Ang2, HIF-1α,VEGF protein in Hp-L positive cases was92.7%,95.1%,90.2%. The data was higher than the Hp-L negative samples (positive rate was31.6%,47.4%,26.3%),the difference had statistical significance (P<0.05), the infection ofHp-L was positively related with the expression of Ang2, HIF-1α,VEGF (r=0.643, r=0.555, r=0.649, P<0.05).(4) RT-PCR showed that the expression of Ang2mRNA in gastric cancer was obviously up-regulated than the control group (P<0.05), and it wascorrelated with histological differentiation,lymph node metastasis and TNMstage(P<0.05).Conclusion:1. Ang2,HIF-1α,VEGF were all highly expressed in gastric cancer, and maycollaboratively promote its angiogenesis.2. The infection of Hp-L was one of the important factors of invasion and metastasis ingastric cancer, the mechanism was relevant about the high expression of Ang2, HIF-1α,VEGF in gastric cancer cells.3. Ang2involved in the formation of new blood vessels in genetic level, and it was animportant promoting factor.of infiltration, transfer in gastric cancer. |
