Objective: To investigate the expression and clinical significance of SLC andCCR7,as well as research the corelationship of the SLC and CCR7with MMP-9andHelicobacter pylori L-type for exploring mechanisms in the gastric cancer.Methods:(1) Collected60cases of gastric cancer surgical specimens of paraffinblocks and clinical pathological data were obtained from cancer weeks (at thejunction of normal and cancer) tissues (30cases, as a cancer week group),44cases ofperipheral tissues (as normal control group). Immunohistochemistry (Elivisionmethod) tissue detected by the SLC and its receptor CCR7and MMP-9proteinexpression.(2) Using Reverse transcription polymerase chain reaction method(RT-PCR) detected30cases of fresh gastric carcinoma and peripheral tissues (ascontrols) in the SLC mRNA expression.(3) The Gram stain and Immunohistochemist-ry were used to detect the60cases of paraffin tissue in the Hp-L-type infection.Results:(1) SLC test results showed that:①immunohistochemical results, thecontrol group was significantly higher than that of SLC gastric cancer group (P<0.05),and SLC expression in cancer tissues with lymph node metastasis (P<0.05), andgender, age, differentiation, invasion depth, and independent of clinical stage (allP>0.05).②RT-PCR showed that expression of the control group SLCmRNAsignificantly increased compared with gastric cancer (P<0.05), and SLCmRNAexpression and lymph node metastasis (P<0.05), and sex, age, histological grade,depth of invasion and clinical stage unrelated (all P>0.05).(2) CCR7results in thecontrol group, CCR7expression was significantly lower than the gastric cancer groupand the adjacent group (P<0.05), and gastric cancer group and the adjacent group ofpositive rate was no significant difference (P>0.05). CCR7expression in the cancergroup was no significant different with sex, age, degree of (P>0.05), but with depth of invasion, lymph node metastasis and clinical stage (all P<0.05).(3) MMP-9testresults showed that the control group, MMP-9expression was significantly lowerthan the gastric cancer group and the adjacent group (P<0.05), gastric cancer andadjacent The positive expression rate showed no significant difference (P>0.05),MMP-9expression in the cancer group with sex, age, degree of differentiation(P>0.05), but not with the depth of tumor invasion, lymph node metastasis andclinical stage (P<0.05).(4) of SLC and its receptor CCR7and MMP-9found that thecorrelation analysis, CCR7and MMP-9was a significant positive correlation between(r=0.748, P<0.05), SLC and no correlation between MMP-9resistance (r=0.006,P>0.05).(5) Hp-L-type test results, Gram staining, immunohistochemical detection oftwo methods results of Hp-L-type with the same (P<0.05), Hp-L-type in gastriccancer group and normal adjacent group The positive rate was68.3%,60.0%,22.7%,gastric cancer, compared with the control group was statistically significant (P<0.05),adjacent compared with the control group are statistically significant (P<0.05),adjacent gastric cancer compared with no significant (P>0.05). The positive rate ofHp-L infection in the cancer group is independent with sex, age, histological grade,clinical stage (P>0.05), but not with depth of invasion, lymph node metastasis (allP<0.05).(6), Hp-L-type gastric cancer patients positive for SLC protein positive rate(9.8%), lower than the Hp-L-type negative group (26.3%), the difference was notsignificant (P>0.05). SLC expression of Hp-L no correlation between infection(r=0.216, P>0.05). Hp-L-type gastric cancer with positive expression of CCR7positive rate (97.6%), higher than the Hp-L-type negative group (42.1%), thedifference was statistically significant (P<0.05). The Spearman rank correlationanalysis test showed that, CCR7expression and Hp-L infection was a significantpositive correlation between (r=0.645, P<0.05).Conclusion:1. SLC may be related to gastric cancer, lymph node metastasis.2. CCR7expression in gastric cancer and gastric cancer and depth of invasion,pathological stage and lymph node metastasis, the incidence of gastric cancer and itsexpression may play a role in promoting development. 3. CCR7and MMP-9expression in gastric cancer was significantly correlated, mayconform to promote invasion and metastasis of gastric cancer.4. Hp-L-type infection is the invasion and metastasis of gastric cancer is oneimportant factor, and the mechanism of gastric cancer cells the expression of CCR7upregulation.
