Relations Between The Expression Of Terc And P16ink4, Ki67in The Diagnosis Of Invasive Cervical Adenocarcinoma | | Posted on:2013-05-06 | Degree:Master | Type:Thesis | | Country:China | Candidate:S Liu | Full Text:PDF | | GTID:2234330374966336 | Subject:Obstetrics and gynecology | | Abstract/Summary: | PDF Full Text Request | | Objectiveâ‘ To detect the human telomerase gene(TERC) amplification fordualcol or interphase DNA probe fluorescence in situ hybridizati on (FISH) ofcervical carcinoma and CIN.â‘¡To investigate the expression, biologicalsignificance and value of P16INK4Aand Ki-67in the diagnosis of the invasivecervical adenocarcinoma.â‘¢Relations between the expression of TERC andpl6ink4,ki67in the diagnosis of Invasive Cervical AdenocarcinomaMethodsâ‘ 195cases of cervical lesion were detected TERC expression byFISH.â‘¡The immunohistochemical SP was used to detect the expression of theP16INK4Aand Ki-67proteins of95cases. For the34cases of normal Cervicalglandular epithelium as control and the other61cases are cervicaladenoeareinoma patients. Fromowitz method was carried out to analysis P16INK4Aand Ki-67in qualitative and semi-quantitatively determination, Clinical andpathologic data of61cases of invasive cervical adenocarcinoma wereretrospectively analyzed.â‘¢Combination of FISH to detect cervical tissue TERCgene amplification and p16ink4and Ki-67expression analysis of early diagnosisand prognosis of cervical adenocarcinoma.Resultsâ‘ Among195cases,the positive rate of TERC gene amplification inChronic cervicitisã€CI Nâ… ã€CI Nâ…¡-â…¢ã€cervical squamous cell carcinomaã€cervica1adenoeareinoma was respectively3.03%(1/33)ã€29.41%(10/34)ã€72.97%(27/37)ã€100%(30/30)ã€91.8%(5/61);there was statistical difference betweenresults of FISH for TERC on cervical squamous cell carcinoma and cervica1adenoeareinoma with the Chronic cervicitis and CIN(P<0.05)②In the positiveexpression of the P16INK4Aand Ki-67in the Normal glandular epithelium was23.53%,20.59%,the positive expression of the p16ink4and Ki-67in the cervicaladenoeareinoma was80.33%,81.97%(P<0.05);the expression level of P16INK4Aincervical adenoeareinoma was related the clinical stage, cervical stromal invasion depth and the diameter of tumor, and was irrelevant to pathological types, tumormalignancy and lymph node metastasis(P>0.05). The positive expression of Ki-67in the cervical adenoeareinoma was related to histologicol stage, the diameter oftumor and lymph node metastasis(P<0.05). The expression of P16was positivelycorrelated to Ki-67in cervica1adenoeareinoma(P<0.01).â‘¢The expression ofP16and Ki-67was positively correlated to the positive rate of TERC geneamplification in cervica1adenoeareinomaConclusionâ‘ the positive rate of TERC gene amplification TERC has becomeincreasing more serious comply with cervixc disease, there is no diferencebetween cervical squamous cell carcinoma and cervica1adenoeareinomaï¼›thedetection of amplification of TERC by FISH can assist in identifyingcervicallesions, and has high Sensitivity and specificity。②The positive expression of thep16ink4and Ki-67in the Normal glandular epithelium is significantly lower thanthe cervical adenoeareinoma. By all appearance, investigate the positiveexpression of p16ink4protein and Ki-67proteins in cervica1adenoeareinomahave significantly clinical value in diagnosis of cervica1adenoeareinoma.â‘¢theImmunohistochemical detection for p16〠Ki-67protein and of amplification ofTERC by FISH can assist in identifying cervical lesions, epeciallyadenocarcinoma.and has high Sensitivity and specificity。... | | Keywords/Search Tags: | FISH, CIN, TERC, p16ink4protein, Ki-67protein, CervicalAdenocarcinoma | PDF Full Text Request | Related items |
