Objective:Colorectal cancer is one of the common malignant gastrointestinal cancers, which is a serious hazard to human health. Currently the disease incidence is rising in some big cities in China. The incidence, development, invasion and metastasis of tumor is a very complex process, has been carried out extensive research, but its exact mechanism is not yet clear.The study through detect the expression of matrix metalloproteinase-9(MMP-9) and smooth muscle22a (SM22α) protein in colorectal cancer tissues and normal tissue, combined with clinical pathologic data to analysis the relationship between this proteins and colorectal cancer occurrence, development, invaded, transfer.Methods:51cases of colorectal carcinoma and normal colorectal tissue samples were collected at the Fourth Hospital of Hebei Medical University department of Surgery2, July2010to December2011. The expression of MMP-9and SM22a was determined in30cases of tissue samples by immunohistochemical S-P method; the other21cases of tissue samples were determined by Western blot. To study the differences of MMP-9and SM22a in cancer and normal tissue, Dukes stage, lymph node metastasis and invasion depth.Results:1The expression level of MMP-9and SM22a in cancer tissue and normal tissueThe immunohistochemical detection results showed that,77.78%of the patients (21/27) of MMP-9expression levels in cancer tissue was much higher than those in normal tissue (P<0.01); the SM22a expression levels of82.14%of the patients(23/28) in cancer tissue was less than in normal tissue (P<0.01). Application Western blot analysis the expression levels of MMP-9and SM22α,95.24%of the patients (20/21) of MMP-9expression levels in cancer tissue was much higher than those in normal tissue (P<0.01); the SM22a expression levels of82.14%of the patients (17/21) in cancer tissue was less than in normal tissue(P<0.01). The MMP-9expression and SM22a expression has a negative correlation in colorectal tumor formation.2The correlation between MMP-9and SM22a expression and the clinicopathologic features in CRCThe result of immunohistochemical showed that, the expression levels of MMP-9was no correlated with Dukes stage (Dukes B and Dukes C), lymph node metastasis, invasion depth (without tumor infiltration serous and infiltrating) and different tumor sites (colon and rectum)(P>0.05). The discovery has also been confirmed by Western blot.Immunohistochemistry and Western blot analysis showed that, the expression levels of SM22a was no correlated with Dukes stage (Dukes B and Dukes C), lymph node metastasis, and different tumor sites (colon and rectum)(P>0.05). And Immunohistochemistry results showed that there was no correlation between SM22a expression and the invasion depth (without tumor infiltration serous and infiltrating).3The correlation between MMP-9and SM22a expressionThe immunohistochemical results of25patients showed that, MMP-9high expression and SM22a low expression was two cases, MMP-9low expression and SM22a high expression was four cases, both high expression is18cases. The MMP-9and SM22a expression has a negative correlation, but there was no statistical significance (r=-0.076, P=0.718). The result is similar by Western blot (r=-0.038, P=0.871).Conclusion:1The expression levels of MMP-9is higher in colorectal cancer tissues than in normal colorectal tissue, and the expression levels of SM22a is less.2There is no correlation between the expression levels of MMP-9, SM22a and Dukes stage (Dukes B and Dukes C), infiltration depth, lymph node metastasis and tumor location.3There is no correlation between MMP-9and SM22a expression in colorectal cancer.
