ObjectiveObstructive jaundice is a set of common symptom.Because of the narrow or stenosis of intrahepatic capillary bile duct, small bile duct, hepatic duct or common bile duct, the bile can’t get through the bile duct into the duodenum but palirrhea into the blood. A lot of diseases can lead to obstructive jaundice, such as the stone of bile duct, bile duct parasite disease, bile duct congenital anomaly, sclerosing cholangitis, tumor of primary in hepato, bile duct, pancreas, etc.The body’s immune function of obstructive jaundice is damaged. Obstructive jaundice causes humoral immune function obstacle and cellular immune function obstacle, and cellular immune function obstacle is more important. The mechanism is closely related to high bilirubin, high bile acid, endotoxemia, inflammatory mediator and so on. For obstructive jaundice the important treatments are as follows:adopting effective measures to remove obstruction, drug intervention, improving the patient’s immunity, preventing the occurrence of complications and reducing mortality rates, and they are the important research subjects of obstructive jaundice.This study observes protective action for immune system after using Danhong injection intervention of obstructive jaundice patient. There is no report related Danhong injection to rat’s obstructive jaundice immune system research. This study applies Danhong injection interventing the rat model of obstructive jaundice, to discuss Danhong’s protection function and mechanism about obstructive jaundice immune system, and provide a new method for clinical treatment of obstructive jaundice.This study determines transformation of CD4, CD8, the serum alanine amino shift enzyme (ALT), the nmda amino shift enzyme (AST) and the total bilirubin (TBIL), the total bile acid of experimental rat, and carry on statistics to learn processing, and observe the morphological change of hepatic tissue, to discuss the immune system protection function and mechanism of obstructive jaundice with Danhong.Methods45female SD rats were divided into three groups at random:(A) sham-operated group:The abdomen of rats was open, but the choledoch was dissociated but not ligation.(B) saline matched group:The abdomen of rats was open, and the choledoch was dissociated, then double ligation with0silk z thread. The beginning of postoperative three day, the1ml saline was injected in the abdominal cavity every day for two weeks.(C) Danhong experimental group:surgical method just as group B, the third day after operation,1ml Danhong was injected in the abdominal cavity every day for two weeks. After operation, all the normal rats were fed, after injection14days all animals were taken blood in vein and heart cavity, and detected CD4, CD8, ALT, AST, TBIL and TBA. The samples were taken hepatic tissue, and conducted at conventional HE dyeing and observed morphological pathology.Results1. Serum CD4, CD8changes:the contents of serum CD4in the matched group was lower than that of sham-operated group and the experimental group, and the matched group serum CDS content was higher than sham-operated group and the experimental group, a statistically significant difference (P<0.05). Serum CD4, CD8numbers approached between the sham-operated group and the experimental group, and they had no statistically significant difference (P>0.05). The CD4content of the experimental group was slightly higher than the sham-operated group, and the CDS content was slightly lower than sham-operated group.2. Serum TBIL,TBA active changes:the serum TBIL of the sham-operated group was obviously lower than ones of the matched group and the experimental group, a statistically significant difference (P<0.05). The serum TBIL content of the experimental group was also obviously lower than those of the matched group, the difference was statistically significant (P<0.05). The serum TBA of the sham-operated group was obviously lower than those of the matched group and the experimental group, a statistically significant difference (P<0.05). The serum TBA of the experimental group was obviously lower than that of the matched group, the difference was statistically significant (P<0.05).3. Serum ALT and AST changes in activity:the ALT, AST of the sham-operated group were obviously lower than those of the matched group and the experimental group, a statistically significant difference (P<0.05). The serum ALT, AST of the experimental group were obviously lower than those of the control group, the difference was statistically significant (P<0.05).4. Pathology morphological changes of hepatic tissue:the liver cells of the sham-operated group were mild edema. The cords of liver cells and the portal area had a few lymphocytes, and the proliferation of the small bile duct was not obvious. The necrosis of liver cells of the matched group was obvious, the cords of liver cells were disorder. There were sporadic liver cells in hyperplasia of fibroblast, proliferation of the small bile duct was obvious. The necrosis of liver cells of the experimental group was lower than the matched group, and there had hyperplasia of fibroblast cells of the cords of liver cells, and the proliferation of the small bile duct was not obvious. The portal area had a few lymphocytes.Conclusion1. Danhong can regulate expression of CD4, CDS and protect T lymphocytes immune that was the damaged by obstructive jaundice.2. The protection of Danhong in obstructive jaundice T lymphocytes immune injury comes true probably by lowing high TBIL and TBA, reducing liver damage.
