In this article,comprehensive extraction conditions about flavone andpolysaccharide in fermented Ampelopsis grossedentata(following the same) isoptimized by response surface analysis; using analysis technology and method aboutchromatography, spectroscopy and mass spectrometry instrument to study of theflavones composition in Ampelopsis grossedentata;basing on serum pharmacologymethod to study of the mechanism of drug action in mice about flavone in Ampelopsisgrossedentata,the main results are as follows:1Optimization of comprehensive extraction conditions about flavoneand polysaccharide in Ampelopsis grossedentataThe effects of extraction temperature,time and ratio of solid to liquid onextraction yields of flavone and polysaccharide from Ampelopsis grossedentata aredetermined individually.Selecting experimental factors and levels base on the resultsof single factor experiment and uses three factors and three levels of response surfaceanalysis method according to a central composite experimental design principle(Box-Benhnken) to optimize of the extraction process.The results show that theoptimal conditions for the extraction of flavone and polysaccharide in Ampelopsisgrossedentata are as follow: temperature95.5℃,time1.63h, solid-liquidratio1:21.05.Under the optimal conditions,the total flavonoids extraction yield was22.49%,the polysaccharide extraction yield was2.03%.2Component analysis about the total flavone in AmpelopsisgrossedentataApplication of UV-Vis scanning, HPLC and LC-MS detection methods for theanalysis of flavonoid components extract from Ampelopsis grossedentata with80%ethanol,the results show: it is detected five compounds in the test range,respectivelycharacterized as3-dihydroxy quercetin, dihydromyricetin,iso-dihydromyricetin,Myricetin-3-O-rhamnoside,dihydroquercetin.Among them,3-dihydroxy quercetin andiso-dihydromyricetin are found in the plant for the first time, which may be associatedwith the fermentation of raw materials.3The study of antioxidant activity of flavone in Ampelopsisgrossedentata in miceUsing serum pharmacology method to investigate the scavenging rate of serumon DPPH radical. It is in order to study of " time-effect relationship" and" dose-effect relationship" about flavone of Ampelopsis grossedentata. the results show: flavone ofAmpelopsis grossedentata enhances the antioxidant capacity of mice;it benefits bestin four hour.In the test range,it is directly proportional to the dose andeffect.Application of HPLC and LC-MS to analysis of mouse serum,thedihydromyricetin is transformed into an electron carrier named forHDF(5,4,7,3’,4’,5’-hexahydroxy-2,3-didehydrogen flavanone) after digestion andabsorption.HDF can receive positive ions and reduce the content of free radicals. Itmay be relate to antioxidant mechanism of action about flavone of Ampelopsisgrossedentata.
